摘要
目的探讨水通道蛋白8(aquaporin 8,AQP8)与结肠癌细胞侵袭转移的关系。方法应用免疫组组织化学链霉卵白素-过氧化物酶连结(SP)法检查18例发生转移的结肠癌、15例未发生转移的结肠癌原发肿瘤组织中AQP8的表达水平差异;结肠癌细胞系Caco-2细胞中过表达AQP8后聚丙烯酰胺凝胶电泳法检测上皮细胞间充质转化(epithelial mesenchymal transition,EMT)相关蛋白(钙黏蛋白和波形蛋白);Transwell侵袭实验检测过表达AQP8后细胞的侵袭能力,定量分析穿膜细胞数量。结果 AQP8在发生转移的结肠癌组织中高表达;AQP8能促进EMT的发生,细胞表型由上皮细胞向间质细胞转化;Transwell实验显示,过表达AQP8后肿瘤细胞穿膜数为88.0±7.2,显著高于对照组(51.6±4.0;P<0.01)。结论 AQP8通过促进EMT的发生,促进结肠癌细胞侵袭转移。
Objective To investigate the role of aquaporin 8 (AQP8) in invasion and metastasis of colon cancer cells. Methods The expression of AQP8 was detected using iramunohistochemical stai- ning with streptavidin peroxidase in colon cancer tissues (cancer with metastasis, n = 18; cancer with- out metastasis,n = 15). After overexpression of AQP8 in Caco-2 cells, epithelial mesenchymal transi- tion (EMT) related protein (E-Cadherin and Vimentin) was detected by polyacrylamide gel electro- phoresis method. The ability of cell invasion was tested using Transwell chamber invasion model, and the number of transmembrane cells was quantitatively analyzed. Results The AQP8 expression was increased in colon cancer tissues. AQP8 enhanced EMT, and cell phenotype changed from epithelial to mesnchyrnal cells. Overexpression of AQP8 could increase the invasion ability of Caco-2 cells. Conclu- sions AQP8 enhances invasion and metastasis of colon cancer cells through EMT.
