Septin-9在子宫内膜癌组织中的表达及意义

Expression and significance of Septin-9 in endometrial carcinoma

目的探讨Septin-9在子宫内膜癌组织中表达及及意义。方法选取子宫内膜癌患者63例、子宫内膜不典型增生患者30例和正常子宫内膜组织42例作为对照组,利用实时荧光定量PCR技术检测不同组织中Septin-9基因表达,所有子宫内膜癌患者术后均获得随访,随访截止日期2015年4月30日,以子宫内膜癌组织中Septin-9 mRNA表达量的中位数为界值,将患者分为高表达组和低表达组,利用Kaplan-Meier生存分析法分析不同Septin-9表达水平对子宫内膜癌患者术后生存时间的影响。结果子宫内膜癌组织中Septin-9 mRNA表达量高于子宫内膜不典型增生组织和对照组,且子宫内膜不典型增生组织高于对照组,差异均有统计学意义(F=29.673,P<0.05);子宫内膜癌组织中Septin-9 mRNA表达量与组织学分级、FIGO分期和淋巴结转移有关(t值分别为17.921、5.211和6.042,P<0.05);Kaplan-Meier生存分析显示,低表达组患者术后平均生存时间77.4个月,高表达组患者术后平均生存时间59.0个月,Log Rank检验显示两组患者术后生存时间差异有统计学意义(χ~2=5.238,P=0.022)。结论 Septin-9在子宫内膜癌组织中呈高表达,可能在子宫内膜癌发生、进展过程中发挥重要作用,且与患者预后密切相关。

Objective To investigate the expression and clinical significance of Septin-9 in endometrial carcinoma. Methods Totally 63 cases of endometrial carcinoma,30 cases of endometrial atypical hyperplasia and 42 cases of normal endometrial tissues in control group were selected. The expressions of Septin-9 in different tissues were detected by using real-time fluorescence quantitative PCR. All endometrial cancer patients after surgery were followed up. Follow-up deadline was April 30,2015. The median value of the expression level of Septin-9 mRNA in endometrial carcinoma tissue was taken as the community value,and the patients were divided into high expression group and low expression group. The effects of different levels of expressions of Septin-9 in endometrial carcinoma tissues on the survival time after surgery were analyzed by using Kaplan-Meier survival analysis. Results The expression levels of Septin-9 mRNA in endometrial carcinoma tissues were higher than in atypical endometrial hyperplasia tissues and in the control group,and those in atypical endometrial hyperplasia tissues were higher than in the control group. The differences were statistically significant（ F = 29. 673,P〈0. 05）. The expression levels of Septin-9 mRNA in endometrial carcinoma tissues were correlated with histological grade,FIGO stage and lymph node metastasis（ t value was 17. 921,5. 211 and 6. 042,respectively,all P〈0. 05）. Kaplan-Meier survival analysis showed that the average survival time of patients in low expression group was 77. 4 months and 59. 0 months in high expression group. Log Rank test showed the difference in the survival time between two groups after surgery was statistically significant（ χ^2= 5. 238,P = 0. 022）.Conclusion Septin-9 is highly expressed in endometrial carcinoma. It might play an important role in the occurrence and progression of endometrial carcinoma,and it is closely related to the prognosis of patients.
