摘要
目的建立测定山药中薯蓣皂苷含量的反相高效液相色潽法(RP-HPLC法)。方法山药粉末经70%甲醇浸泡过夜,回流提取1h,采用RP-HPLC法进行测定。色谱柱:大连伊力特SinoChrom ODS-BP色谱柱(250mm×4.6mm,5μm),流动相:甲醇-水(84∶16),流速:1 mL/min,检测波长:206nm,柱温:30℃。结果回归方程为:Y=7 446 X-22 054,线性范围:17.0~40.8μg/mL(r=0.999 2),分离度>1.5,回收率为102.14%,RSD为1.76%,铁棍山药、面山药、有机山药中薯蓣皂苷含量分别为0.504 7、0.420 5、0.649 6mg/g。结论该方法简便、准确、可靠,可用于山药中薯蓣皂苷的含量测定。
Objective To establish a RP-HPLC method to determine the diosgenin content in Rhizoma Dioscoreae.Methods Yam powder was soaked overnight by 70% methanol,and refluxed samples for 1h,and measured by RP-HPLC method.Column Dalian Iraqi special forces Sino Chrom ODS-BP column(250mm×4.6mm,5μm),mobile phase:methanol-water(84∶16),flow rate:1.0mL/min,deteceion wavelight:206nm,temperature:30℃.Results The regression equation was:Y=7 446 X-22 054,linear range was 17-40.8g/mL(r =0.999 2),separation degree〉1.5,recovery rate〉102.14%,RSD was1.76%.The organic content of dioscin in yam were:0.504 7,0.420 5,0.649 6 mg/g.Conclusion The method is simple,accurate and reliable,which can be used for the determination of diosgenin in yam.
