摘要
以美国进口玉米酒糟粕(DDGS)中转基因玉米品系MIR162的实时荧光PCR检测方法为研究内容,分别采用3种常见商业化试剂盒提取基因组DNA,采用3个检测标准中规定的实时荧光PCR方法进行MIR162定性检测,比较了9种方法组合在检测玉米酒糟粕中转基因成分的检测技术性能。结果表明,使用TIANGEN?植物基因组DNA提取试剂盒和《SN/T 1196—2012转基因成分检测玉米检测方法》规定的荧光PCR方法,检测灵敏度最高,符合现行检验监管法规要求。考虑到大宗农产品及其加工产品的国际贸易普遍存在转基因成分低水平混杂的情况,进一步探讨了进口农产品转基因成分检验监管分类管理的必要性,提出针对不同风险级别的进口农产品采用灵敏度相适宜的检测方法。
By real-time fluorescence PCR methods for detection of GM maize event MIR162 in distillers dried grains with soluble (DDGS) exported from US, 3 universal commercial kits were used for genomic DNA extraction, and 3 real-time fluorescencePCR methods provided in detection standards were applied for the qualitative detection of MIR162. A comparison of the 9 detection combinations was made to investigate the technical performances of GM ingredient detection for DDGS. The results showed that the combination of TIANGEN plant genomic DNA extraction kit and the real-time fluorescence PCR method in" SN/T 1196- 2012 detection of genetically modified components- maize test methods" gave a better detection sensitivity to satisfy the requirements of current inspection and supervision rules. Based on the widespread existence of low level presence (LLP) of GM ingredients in international trades for bulk agricultural products and their processed products, we further discussed the necessity of classified inspection and supervision on GM ingredients in the imported agricultural products, and also presented the application of detection methods with specific sensitivities which suitable for imported agricultural products at different risk levels.
