摘要
本研究旨在研究太行山羊MC1R基因在不同毛色皮肤组织中的表达规律。运用RT-PCR方法从纯黑色太行山羊皮肤组织中扩增了MC1R基因的编码区序列,运用生物信息学方法分析了其编码蛋白的结构特点,利用实时荧光定量PCR技术,对4种不同毛色(纯黑、纯白、黄褐、黑斑白)各4只太行山羊,共计16个个体的皮肤组织进行mRNA表达研究。结果表明:MC1R基因CDS区长954 bp,编码317个氨基酸,其编码蛋白是G蛋白偶联受体家族的成员;同源性比对和进化树结果显示,与绵羊、家牛和水牛同源性较高;荧光定量结果显示,MC1 RmRNA在纯黑色个体皮肤组织中表达量最高,黄褐色次之。本研究结果为进一步研究该基因在毛色形成中的作用以及对纯黑色太行山羊的纯繁选育奠定基础。
This study aims to research the m RNA expression rules of Tai-hang goat MC1R gene in skin tissues with different colors. The coding sequence(CDS) of MC1R gene was amplified from skin tissues of Tai-hang goat with pure black color using reverse transcription polymerase chain reaction(RT-PCR). Structural characteristics of coded protein were analyzed by bioinformatics methods. Sixteen Tai-hang goats with four kinds of coat colors(black, white, brown and white with black-spots, respectively) were chosen to study the mRNA expression in skin tissues using quantitative realtime PCR. Results showed that the length of complete CDS was 954 bp encoding a protein with 317 amino acids, and the encoded protein is a member of G protein-coupled receptor family. Results of homology alignment and evolutionary tree showed its high homologies with sheep, cattle and buffalo. MC1R m RNA showed the highest expression in pure black skin tissues, followed by that in brown skin tissues. These results laid a foundation for further study on the role of MC1R gene in the formation of coat colors, and the pure breeding of black Tai-hang goat.
