摘要
目的:观察三七总皂苷(panax notoginseng saponins,PNS)对拟缺血损伤脑微血管内皮细胞胞浆型磷脂酶A2IVA(phospholipase A2 group IVA,PLA2G4A)表达及其功能的影响。方法:培养大鼠原代脑微血管内皮细胞,氧糖剥夺法制备拟缺血模型,用三七总皂苷及PLA2G4A抑制剂进行干预,通过RT-PCR和Western blotting法检测各组PLA2G4A基因及蛋白表达水平,ELISA法检测各组花生四烯酸(arachidonic acid,AA)、20-羟二十烷四烯酸(20-hydroxyeicosatetraenoic acid,20-HETE)、血栓素A2(thromboxanea2,TXA2)/前列环素(prostaglandin I2,PGI2)表达水平。结果:三七总皂苷可显著下调PLA2G4A的基因和蛋白表达,对AA、20-HETE、TXA2/PGI2也有下调作用。结论:三七总皂苷可以下调拟缺血损伤脑微血管内皮细胞PLA2G4A及其下游产物的表达,这可能是三七总皂苷在脑缺血损伤中发挥化瘀通络作用的机制之一。
Objective to evaluate the effects of Panax Notoginseng Saponins (PNS) on the PLA2G4A in Mimic isehemia brain microvascular endothelial cells (BMECs). Methods: primary rat BMECs were used and mimic cerebral isehemia in vitro was established by oxygen-glucose-deprived (OGD)method. OGD-induced BMECs were treated with PNS and Pyrrophenone(PLA2C,4A inhibitor)respectively. The expression of PLA2G4A in mRNA and protein were detected by RT-PCR and Western blot methods. The expression of AA 20-HETE TXA2/PGI2 were detected by Elisa method. Results : both mRNA and protein expression of PLA2C,4A in OGD-induced BMECs were down-regulated by PNS. AA 20-HETE TXA2/PGI2 were also decreased in administration group. Conclusion:PNS can down regulate the expression and function of PLA2G4A,which may be the internal mechanism of PNS's Anticoagulant effect in cerebral ischemia.
