淫羊藿苷元微乳制剂对小鼠免疫性血小板减少性紫癜模型的治疗作用 被引量:11

Effects of anhydroicaritin microemulsion formulation on the immunologic bocytopenic purpura mice

摘要 目的:研究淫羊藿苷元对小鼠免疫性血小板减少性紫癜(immunologic thrombocytopenic purpura,ITP)模型的治疗作用。方法:雄性BALB/c小鼠随机分为5组,除正常对照组外,从给药d 1开始至d 9,小鼠每隔1 d腹腔注射豚鼠抗小鼠血小板血清(1∶4,100μL)诱导小鼠长期ITP。分离BALB/c小鼠血小板皮下注射至豚鼠体内,连续5周,制备豚鼠抗小鼠血小板血清。4组ITP模型小鼠分别灌胃(ig)给药10 d:微乳基质、重组人血小板生成素(recombinant human thrombopoietin,rh TPO)3 000 U·kg-1、淫羊藿苷元微乳制剂(Y003:1.25和5.0 mg·kg-1),空白对照组给予微乳基质。d 11处死动物,取血、股骨、脾脏,并测定以下指标:血常规、脏体比、流式网织血小板计数、ELISA测定血清中血小板生成素(thrombopoietin,TPO)含量、脾脏病理学检测。结果:1.25 mg·kg-1剂量给药组与模型组比较,外周血小板数、网织血小板比例、脏体比、血清TPO含量等指标均有显著差异(P<0.05)。结论:口服淫羊藿苷元微乳制剂能有效治疗小鼠ITP模型血小板减少症状,其作用机制可能与激动C-Mpl受体刺激血小板增殖分化有关。 Objective: To investigate effects of anhydroicaritin microemulsion formulation on the immunologic thrombocytopenic purpura( ITP) mice. Methods: Male BALB/c mice were randomly divided into five groups.Constant ITP was induced in four groups by intraperitoneally( ip) injecting guinea pig anti-mouse thrombocyte antiserum with a dose of 100 μL at 1 ∶ 4 dilution every other day,and drug treatment was given every day at the same time. The control group was ip injected with equal volumes of saline. The four groups of the thrombocytopenia mice were treated with microemulsion matrix,rh TPO 3 000 U·kg-1,drug Y003 1. 25 mg·kg^-1( Y003-L),and drug Y003 5. 0 mg·kg^-1( Y003-H),respectively. The control group was treated with microemulsion matrix. After eleven days of treatment,the blood and spleens were taken and the following indices were determined: complete blood count,organ-to-body weight ratio,multiparameter flow cytometry reticulated platelets counts,TPO content in peripheral blood by ELISA test,and histopathology test of spleen. Results: The platelet counts of peripheral blood of groups Y003-L and Y003-H were significantly increased( P〈0. 05). Reticulated platelets counts,organ-to-body weight ratio,and TPO content in the peripheral blood of group Y003-L were significantly decreased( P〈0. 05). Conclusion: Oral anhydroicaritin microemulsion formulation can improve the symptoms of thrombocytopenia in ITP mice effectively. The mechanism of anhydroicaritin microemulsion's beneficial effect may be related to the C-Mpl receptor-mediated stimulation of proliferation and differentiation of platelets.
