摘要
冷冻保存后的GV期卵母细胞内大量积累ROS,影响胚胎发育。NAC作为一种抗氧化剂,可缓解氧应激造成的损伤。但NAC对冷冻GV期卵母细胞是否存在维持作用尚待研究。为探究NAC在冷冻、解冻后小鼠GV期卵母细胞发育中作用,检测冷冻保存后小鼠GV期卵母细胞在添加不同浓度NAC成熟培养液后成熟情况,观察其卵裂率与囊胚率、线粒体分布、ROS水平和受精后胚胎发育等。结果表明,添加1.5 mmol·L^(-1)NAC,冷冻保存后,小鼠GV期卵母细胞,线粒体排布均匀,卵母细胞比例显著提高,ROS水平降低。NAC可改善小鼠GV期卵母细胞经冷冻保存后复苏和体外成熟水平,提高其发育能力。
Large amounts of ROS were accumulated in frozen-thawed GV-stage oocytes, which could affect embryonic development. NAC, as an antioxidant, could alleviate the damage caused by oxidative stress. However, currently, it remained to be seen whether NAC has a maintenance effect in frozen-thawed GV-stage oocytes. To investigate the role of NAC in frozen-thawed mouse GV-stage oocytes development, the maturation of frozen-thawed mouse GV-stage oocytes was examined at maturation medium added different concentrations of NAC and the cleavage rate, blastocyst rate, mitochondrial distribution, ROS level and embryo development after fertilization were observed. The results showed that the mitochondria were homogeneous, the proportion of oocytes was significantly increased, and the level of ROS was decreased into cryopreserved mouse GV-stage oocytes after adding 1.5 mmol. L-1 NAC. These results suggested that NAC can improve the recovery and in vitro maturation of frozen-thawed mouse GV-stage oocytes and improve their developmental potency.
