摘要
为热带地区甘薯引种和新品种培育提供重要的理论依据,通过农艺性状比较筛选出在海南地区适应性较好的30份甘薯引种种质资源,利用SSR分子标记技术对30份甘薯种质资源进行遗传多样性分析,明确其在DNA分子水平上的遗传差异。结果表明:11对SSR引物可扩增出57条清晰的条带,其中多态性条带53条,多态性百分率93.0%,平均每对引物5.18条。经遗传相似性和聚类分析,30个甘薯品种遗传相似性系数在0.42-0.96,平均为0.62,甘薯品种间遗传多样性较丰富。以0.61的相似系数为阈值,30份材料聚成3大类。
To provide a theoretical basis for sweet potato incroduction and new varieties cultivation in tropical region, through assessment of the agronomic traits, the authors selected 30 sweet potato germlines with good adaptability in Hainan area. In this study, the genetic diversity of these 30 sweet potato germlines was analyzed by SSR molecular marker technology, thus to clarify the genetic variation at DNA molecular level. Results: 57 clear and reproducible SSR bands could be recorded according to the electrophoresis results of 11 pairs of SSR primers. Among them, 53 were polymorphic bands, with an average of 5.18 bands per primer pair and the percentage of polymorphism up to 93.0-. the analysis of the genetic similarity and UPGMA clustering showed that coefficients of genetic similarity varied from 0.42to 0.96 with the mean of 0.62, indicating high genetic diversity. Using the genetic similarity value of 0.61 as the threshold,the 30 germlines were clustered into three clades.
