脂肪干细胞覆膜聚丙烯补片减轻疝修补术后炎症反应的研究 被引量:3

摘要 目的探讨脂肪干细胞(ADSCs)覆膜聚丙烯补片对大鼠疝修补术后炎症反应的影响。方法培养扩增检测脂肪干细胞并接种到聚丙烯补片上,体外培养10天。成年SD大鼠12只,分为覆膜组6只,未覆膜组6只,分别将ADSCs覆膜聚丙烯补片和聚丙烯补片植入覆膜组和未覆膜组大鼠的腹壁肌肉表面,检测大鼠术前及术后3、7、14、21、28天血常规、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)水平。结果 ADSCs在聚丙烯补片上附着生长良好,免疫荧光检测显示干细胞特异性标记CD90表达≥97%。覆膜组在术后14天炎症反应高峰时期白细胞计数[(10.86±0.56)×109/L比(11.92±0.56)×109/L,P<0.05]、TNF-α[(87.12±12.59)μg/L比(175.28±34.37)μg/L,P<0.01]、IL-1β[(275.69±54.28)μg/L比(501.36±98.15)μg/L,P<0.01]均低于未覆膜组,差异有统计学意义。结论与聚丙烯补片组比较,ADSCs覆膜聚丙烯补片可减轻大鼠疝修补术后全身炎症反应。 Objective To investigate the effect of adipose-derived stem cells(ADSCs) coated polypropylene patch(PP) on relieving the inflammation after herniorrhaphy. Methods The ADSCs were seeded into polypropylene mesh and in vitro cultured for 10 d. Twelve SD rats were randomly divided into control group and experimental group,with 6 rats in each group. PP coated with ADSCs and PP were implanted into the abdominal wall muscles of experimental group and control group, respectively. The blood samples were collected before and 3, 7, 14, 21 and 28 d after operation to measure white blood cell(WBC) count, TNF-α and IL-1β. Results ADSCs grew well on the polypropylene patch, and the CD90 expression was more than 97% by immunofluorescence. In the peak period of inflammatory reaction on Day 14, the WBC count[(10.86±0.56)×109/L vs(11.92±0.56)×109/L, P〈0.05], TNF-α(87.12±12.59μg/L vs 175.28±34.37μg/L, P〈0.01), IL-1β(275.69±54.28μg/L vs 501.36±98.15μg/L, P〈0.01) in experimental group were lower than those of control group. Conclusion Polypropylene patch coated with ADSCs can effecti-vely reduce the systemic inflammatory response after herniorrhephy in rats.
