益肾生髓法对骨质疏松雌鼠骨密度、骨代谢影响的研究 被引量:1

摘要 目的观察益肾生髓法对骨质疏松(osteoporosis,OP)雌鼠骨密度、骨代谢影响的研究,从生化分子水平探讨OP发病机制及益肾生髓中药防治OP的作用机制。方法清洁级去卵巢成年雌鼠45只,按随机数字表法分为中药干预组、西药对照组、正常对照组,每组各15只,同时分别给予中药汤剂、碳酸钙D3颗粒、生理盐水灌胃给药。16周后取材血清,自动生化分析仪测定血清Ca、P、BGP、BALP浓度,股骨在双能X线骨密度仪测定骨密度,并在显微镜下观察骨组织结构的改变。结果中药干预组及西药对照组用药16周后,与正常对照组比较,血清Ca、P浓度及骨密度(股骨、脊柱及全身)表达水平均明显上调(P<0.05);其中中药干预组BALP及骨密度(股骨、脊柱及全身)上调幅度大于西药对照组,而BGP则大幅减低(P<0.05)。给予西药对照组、中药干预组16周后骨组织形态计量学测定与正常对照组比较均有显著差别,明显高于正常对照组,差异有统计学意义(P<0.05)。结论益肾生髓法对OP雌鼠上调骨密度、血清Ca、P浓度,抑制BGP,改变骨组织结构,从而促进骨形成,对减缓去卵巢雌鼠骨质疏松的发生发展有一定的作用。 Objective To observe the effect of nourishing kidney and marrow method on bone mineral density and bone metabolism in osteoporosis (osteoporosis, OP), and to explore the mechanism of traditional Chinese medicine for preventing and treating OP from the biochemical molecular level. Methods Forty-five adult female rats of clean grade were randomly divided into traditional Chinese medicine group, western medicine control group, normal control group, 15 rats in each group, and were given the decoction of Chinese medicine, D3 of calcium carbonate particles, and saline gavage, respectively. After 16 weeks, the serum levels of Ca, P, BGP and BALP were measured by automatic biochemical analyzer. The bone density of the femur was measured by dual energy X-ray bone density instrument, and the changes of bone tissue structure were observed under microscope. Results Compared with the normal control group, in the traditional Chinese medicine group and western medicine control group after 16 weeks of treatment, serum Ca, P and the density of bone ( femur, spine and whole body) level significantly increased ( P 〈 0. 05 ) ; the traditional Chinese medicine intervention group ( BALP and bone density of femur, spine and whole body) was greater than the western medicine and the control group, while BGP was significantly reduced (P 〈 0. 05 ). While bone tissue morphology measurement of the western medicine control group, traditional Chinese medicine intervention group after 16 weeks compared with normal control group were significantly different (P 〈 0. 05). Conclusion The method of nourishing kidney and marrow increases bone density, serum Ca, P concentration, OP concentration, inhibits BGP, c sue, promotes bone formation, make the occurrence slow down opment of osteoporosis in ovariectomized female rats. hanges the structure of bone tis- and have some effects on development of osteoporosis in ovariectomized female rats.
