摘要
目的卵巢癌是妇科肿瘤中死亡率居首的常见恶性肿瘤,寻找新的生物学标志物以提高卵巢癌诊治的敏感性与特异性至关重要。本研究探讨GTP酶激活蛋白SH3功能区结合蛋白(Ras-GTPase-activating protein SH3domain binding protein,G3BP)和骨桥蛋白(osteopontin,OPN)在上皮性卵巢组织中的表达、意义及其表达的相关性,为卵巢肿瘤的临床诊断及治疗提供依据。方法采用免疫组织化学方法检测G3BP和OPN在来源于中国医科大学盛京医院2009-10-01-2015-05-31的11例正常卵巢组织、20例卵巢上皮性良性肿瘤组织、31例卵巢上皮性交界性肿瘤组织及60例卵巢上皮性恶性肿瘤组织中的表达情况;并对二者表达的相关性进行统计分析。结果 G3BP在正常卵巢上皮组织、卵巢上皮性良性肿瘤组织、卵巢上皮性交界性肿瘤组织及卵巢上皮性恶性肿瘤组织中的表达呈递增趋势,其中上皮性良性肿瘤组织与正常卵巢上皮组织比较,χ2=6.38,P〈0.01;上皮性交界性肿瘤组织与上皮性良性肿瘤组织比较,χ2=1.183,P〈0.01;上皮性恶性肿瘤组织与上皮性交界性肿瘤组织比较,χ2=3.342,P〈0.01。OPN在正常卵巢上皮组织、卵巢上皮性良性肿瘤组织、卵巢上皮性交界性肿瘤组织及卵巢上皮性恶性肿瘤组织中的表达呈递增趋势,其中上皮性良性肿瘤组织与正常卵巢上皮组织比较,χ2=0.005,P〈0.01;上皮性交界性肿瘤组织与上皮性良性肿瘤组织比较,χ2=3.844,P〈0.01;上皮性恶性肿瘤组织与上皮性交界性肿瘤组织比较,χ2=3.429,P〈0.01。G3BP与OPN在卵巢上皮性恶性肿瘤组织中的表达具有正相关性,r=0.351,P〈0.01。结论 G3BP和OPN表达升高与上皮性卵巢癌的发生发展有密切关系,且二者可能存在协同作用。
OBJECTIVE Ovarian cancer is one of the common malignant tumors in female genitalorgans,and its mortality ranks first in gynecdogical tumors.It is essential to find new biomarker for important the sensitivity and specificity of diognosis and treatment of ovarian cancer.The objective of this study was to evaluate the expression level of G3 BP and OPN in different epithelial ovarian tissues,to investigate their correlation in epithelial ovarian cancer,and to provide experimental bases for diagnosis and treatment of ovarian cancer.METHODS The expression of G3BP and OPN in 11 cases of normal ovarian tissues,20 cases of benign epithelial ovarian tumor tissues,31 cases of borderline epithelial ovarian tumor tissues and 60 cases of epithelial ovarian cancer tissues,which were from the patients underwent treatment in Shengjing hospital between 2009-10-1 and 2015-5-31,were detected by immunohistochemistry.And the different expression among those tissues and the correlation between G3 BP and OPN expression were analyzed.RESULTS The expression of G3BP showed an increasing tendency among normal epithelial ovarian tissues,benign epithelial ovarian tumor tissues,borderline epithelial ovarian tumor tissues and epithelial ovarian cancer tissues.The difference were statistically significant(benign epithelial ovarian tumor tissues vs.normal epithelial ovarian tissues,χ2=6.38,P〈0.01);borderline epithelial ovarian tumor tissues vs.benign epithelial ovarian tumor tissues(χ2=1.183,P〈0.01);epithelial ovarian cancer tissues vs.borderline epithelial ovarian tumor tissues(χ2=3.342,P〈0.01).The expression of OPN showed an increasing tendency among normal epithelial ovarian tissues,benign epithelial ovarian tumor tissues,borderline epithelial ovarian tumor tissues and epithelial ovarian cancer tissues;the difference were statistically significant(benign epithelial ovarian tumor tissues vs.normal epithelial ovarian tissues(χ2=0.005,P〈0.01);borderline epithelial ovarian tumor tissues vs.benign epithelial ovarian tumor tissues(χ2=3.844,P〈0.01);epithelial ovarian cancer tissues vs.borderline epithelial ovarian tumor tissues(χ2=3.429,P〈0.01).There was a positive correlation between G3BP and OPN in epithelial ovarian cancer tissues(r=0.351,P〈0.01).CONCLUSION High expressions of G3BP and OPN are involved in tumorigenesis and development of epithelial ovarian cancer,and they may work synergisticlly.
