摘要
目的:探讨宫颈癌中染色体空间重组因子(Rsf-1/HBXAP)的表达及临床意义,为宫颈癌的发生发展机制和靶向治疗提供实验依据。方法:选取2008年12月31日至2014年1月1日在山东省肿瘤医院妇瘤科治疗的患者宫颈组织标本共234例,采用免疫组化SP法检测Rsf-1/HBXAP蛋白在17例正常宫颈组织、35例宫颈上皮内瘤变(CIN)组织和182例宫颈癌组织中的表达,并采用Kaplan-Meier生存分析揭示其与宫颈癌临床病理以及患者预后的关系。应用Real-time PCR技术及Western blot技术检测Rsf-1/HBXAP基因在不同宫颈细胞系中的表达,分析不同细胞系中Rsf-1/HBXAP表达水平差异。结果:正常宫颈组织、CIN组织及宫颈癌组织中Rsf-1/HBXAP蛋白的阳性表达率分别为5.88%(1/17)、37.14%(13/35)及79.12%(144/182),差异有统计学意义(P<0.05)。宫颈癌组织中,Rsf-1/HBXAP蛋白表达与肿瘤分化程度、肿瘤分期及淋巴结转移有关(P<0.05),但与患者年龄、肿瘤大小无关(P>0.05)。He La及Siha细胞系中Rsf-1/HBXAP mRNA、蛋白表达显著高于293T细胞系。Kaplan-Meier生存分析表明,宫颈癌患者中Rsf-1/HBXAP蛋白高表达组其总体生存率显著低于低表达组(P<0.05)。结论:Rsf-1/HBXAP在宫颈癌组织及其细胞系中高度表达,预示其可能参与了宫颈癌的发生和进展,为其作为诊断及判断预后的新的肿瘤标志物以及作为宫颈癌个体化治疗的新靶点提供了依据。
Objective: To investigate the expression and significance of remodeling spacing factor( Rsf-1 / HBXAP) in cervical cancer,and to analyze the gene detection methods and theoretical basis for the cervical cancer targeted treatment. Methods: The immunohisto-chemical staining was used to detect Rsf-1 / HBXAP expression in 17 cases of normal control,35 cases with cervical intraepithelial neoplasm( CIN) and 182 cases with cervical cancer. RNA were extracted from He La,Siha and 293 T by the routine phenol-chloroform method,which were used for PCR amplification. Besides,the expression levels of Rsf-1 / HBXAP in the cell lines were also detected by Western blot.Finally,Kaplan-Meier survival analysis was used to evaluate the difference of overall survival between the high Rsf-1 / HBXAP expression and low Rsf-1 /HBXAP expression.Results: The positive rates of Rsf-1 / HBXAP were 5.88% in normal cervical tissues,37.14% in CIN lesions,79.12% in carcinoma respectively,which was significantly different( P〈0.05). The positive expression rate of Rsf-1 / HBXAP in cervical cancer tissues was correlated with the degree of tumor differentiation and clinical staging( P〈0.05),which was no related to age and size( P〉0.05). Rsf-1 / HBXAP mRNA expression and protein expression in He La and Siha were higher than 293 T.Kaplan-Meier survival analysis revealed that RSF-1 overexpression is significantly related to poor clinical outcome for the women suffering cervical carcinoma.Meanwhile,Fisher ' s exact test and χ-2 test also confirmed these findings. Conclusion:Rsf-1 / HBXAP closely associated with precancerous lesions and cervical cancer,and might provide new information regarding the treatment strategies for cervical cancer combination therapy.
