骨碎补总黄酮对绝经后骨质疏松大鼠疼痛介质和疼痛反应影响 被引量:15

Effect of Drynaria Flavonoids on the Expression of Pain Mediators and Osteoporotic Pain in the Osteoporosis Model of Ovariectomized Rats

摘要 目的:利用去卵巢大鼠骨质疏松模型,观察骨碎补总黄酮对血清中肿瘤坏死因子α(TNF-α)、白介素1(IL-1)、前列腺素E2(PE2)和5羟色胺(5-HT)的表达和对大鼠痛阈的影响。方法:36只雌性大鼠随机分为3组(假手术组、去卵巢组和药物处理组),造模成功后,药物处理组持续给予骨碎补总黄酮,采用ELISA检测大鼠血清中TNF-α、IL-1、PE2和5-HT的表达变化,采用X线骨密度检测仪测定大鼠骨密度;采用von-Frey纤毛检测法观察其不同时间点的镇痛作用。结果:骨碎补总黄酮对去卵巢骨质疏松症导致的骨密度下降有明显抑制作用,且明显抑制大鼠血清中TNF-α、IL-1、PE2和5-HT的表达,显著提高去卵巢骨质疏松大鼠的痛阈。结论:骨碎补总黄酮能明显提高去卵巢大鼠的骨密度,并可抑制血清中疼痛介质的表达,具有镇痛作用。 Objective:To investigate the effect of the drynaria flavonoids on the levels of TNF-α,IL-1,PE2 and 5-HT in the serums and pain threshold by using the osteoporosis model of ovariectomized rat. Methods:Thirty six female virgin Sprague-Dawley rats were randomly divided into 3 groups. After the modeling,drug-treated group was provided with drynaria flavonoids. Bone mineral density was measured by the bone density X-ray measuring instrument and the levels of TNF-α,IL-1,PE2 and 5-HT were assayed by the ELISA. Pain threshold was assessed using von Frey filaments. Results:Compared to the control,bone mineral density,the levels of TNF-α,IL-1,PE2 and 5-HT and pain threshold were significantly increased in the drug-treated group with drynaria flavonoids. Conclusions:Drynaria flavonoids can significantly increase bone density in ovariectomized rats and have an analgesic effect by inhibiting the expression of pain mediators.
