肠炎沙门菌感染诱导鸡肠道自噬发生的研究

Study on Intestinal Autophagy Induced by Salmonella enteritidis Infection in Chickens

试验旨在探究肠炎沙门菌感染诱导鸡肠道自噬发生及其规律。将42只SPF母雏随机分成两组,其中对照组24只,攻毒组18只。5日龄时,攻毒组嗉囊灌服肠炎沙门菌SC070分离株,对照组灌服等体积无菌PBS。于攻毒6、12和24 h(6、12、24 hpi)分别记录雏鸡体重和脾脏重,检测雏鸡肝脏、脾脏、血液、空肠和盲肠组织载菌量,利用透射电镜(TEM)观察雏鸡空肠组织中自噬结构及数量。结果显示,与对照组相比,攻毒组雏鸡在攻毒后6 h即开始检测到脏器、血液及肠道载菌;雏鸡感染肠炎沙门菌后,空肠载菌和盲肠载菌在6、12、24 hpi间均差异极显著(P<0.01),其中空肠载菌在6~24 hpi呈现先减少后增加的规律,而盲肠载菌在6~24 hpi内呈持续降低趋势;攻毒组空肠组织中自噬泡数量明显高于对照组,且在6~24 hpi间呈现先增加后减少的趋势。由此可见,肠炎沙门菌感染能够诱导鸡肠道自噬增加,且在感染后6~24 h内呈先增加后减少的规律。

This experiment was conducted to investigate the intestinal autophagy induced by Salmonella enteritidis infection in chickens. 42 SPF female chicks were randomly divided into two groups,the control group （n=24）and the challenge group （n=18）. At the age of 5 days, the challenge group was perfused with Salmonella enteritidis SC 070 strains by crop, the control group was perfused with same volume of sterile PBS. The body weight （BW） and spleen weight of chicks at 6 hours post injection （hpi）, 12 hpi and 24 hpi were recorded respectively,then the bacteria loads of liver,spleen,blood,jejunum and cecum were deetected. The structure and quantity of autophagy in jejunum were observed by transmission electron microscopy （TEM）. The results showed as follows： Compared with the control group, bacteria were began to detected at 6 hpi in liver, spleen,blood,jejunum and cecum of chicks in the challenge group. The bacteria loads of jejunum and cecum had very significant difference at 6, 12 and 24 hpi （P〈0.01）,and showed a trend of decreasing at first and then increasing in jejunum,while continued decreasing in cecum. The number of autophagic vacuoles in jejunum of the challenge group were significantly higher than that in the control group,showing an increase trend at first and then decrease at 6 to 24 hpi. It was concluded that the infection of Salmonella enteritidis could induce the increase of intestinal tissue autophagy, and showing an increasing trend at first and then decreasing at 6 to 24 hpi.
