摘要
目的探讨潜伏膜蛋白2A(latent membrane protein 2A,LMP2A)、Snail和N-cadherin在鼻咽癌(nasopharyngeal carcinoma,NPC)中的表达及意义。方法通过免疫组织化学检测LMP2A、Snail和N-cadherin在鼻咽黏膜慢性炎、NPC和淋巴结转移灶中的表达,比较3种蛋白的表达相关性及其与临床病理的关系。通过体外实验检测LMP2A对鼻咽癌CNE2细胞形态的影响及Snail和N-cadherin蛋白表达的影响。结果 LMP2A、Snail和N-Cadherin蛋白的高表达与NPC患者临床恶性进展呈正相关(尤其是淋巴结转移);LMP2A与Snail、N-cadherin蛋白表达均呈正相关;体外实验证实LMP2A可上调鼻咽癌CNE2细胞中N-cadherin和Snail蛋白的表达并诱导细胞发生上皮-间质转化(epithelial-mesenchymal transition,EMT)样改变。结论 LMP2A、Snail和N-cadherin在鼻咽癌及其淋巴结转移灶中的高表达与NPC恶性进展和淋巴结转移密切相关。
Objective To investigate the expression and significance of latent membrane protein 2A (LMP2A), Snail and N- cadherin in nasopharyngeal carcinoma (NPC). Methods Immunohistochemistry was performed to detect the expression of LMP2A, Snail and N-cadherin in nasopharyngeal mucosa with chronic inflammation (NCI), NPC and its lymph node metastatic foci ( LNM). The correlations between these proteins, and their relationship with the clinical pathological parameters of NPC were analyzed. The effect of LMP2A on the morphology of nasopharyngeal carcinoma CNE2 cells and the expression of Snail and N-cadherin were detected in vitro. Results The high expression of LMP2A, Snail and N-cadherin was positively related to the malignant progression of NPC, es- pecially lymph node metastasis. The expression of LMP2A was positively correlated with that of Snail and N-cadherin. It was verified in vitro that LMP2A upregulated the expression of N-cadherin and Snail in CNE2 cells and induced changes toward epithelial-mesenchymal transition (EMT). Conclusion The high expression of LMP2A, Snail and N-cadherin in NPC and metastatic lymph nodes is closely re- lated to the malignant progression and lymph node metastasis of NPC.
