摘要
目的:利用香菇多糖注射液对脓毒血症进行干预,研究其保护作用并从肿瘤坏死因子(TNF-α)水平研究其保护机制。方法:应用盲肠结扎穿刺(CLP)法制备脓毒血症动物模型。雄性Wistar大鼠40只,随机分为空白对照组(假手术组)、模型组(脓毒血症组)、香菇多糖治疗组、香菇多糖预防组4组,假手术组和模型组尾静脉推注注射用水0.21ml/kg,连续4周;预防组尾静脉注射香菇多糖注射液0.105mg/kg,1周2次,连续4周后行CLP术;治疗组CLP术后给予香菇多糖注射液静注4周。造模成功及干预后12h采用ELISA法检测各组动物血清中TNF-α的含量。结果:香菇多糖干预后,治疗组和预防组内毒素水平明显低于模型组(P<0.05),模型组PaO_2低于对照组,PaCO_2高于对照组(P<0.05)。预防组PaO_2高于模型组,PaCO_2低于模型组(P<0.05)。模型组、治疗组TNF-α水平高于对照组(P<0.05);预防组、治疗组血清TNF-α水平低于模型组(P<0.05);预防组和治疗组血清TNF-α水平高于对照组(P<0.05)。结论:香菇多糖对脓毒血症有良好的修复和保护作用,且能明显降低脓毒血症大鼠血清中TNF-α的含量,可能是其保护脓毒血症的重要作用机制之一。
Objective:To observe the protective effect of Lentinan injection on tumor necrosis factor of sepsis rats and to explore the mechanism of Lentinan in prevention of sepsis.Methods:Healthy Wistar rats were randomly divided into four groups,control group,experimental group,Lentinan prevention group and Lentinan treatment group.Sepsis models were induced by cecal ligation and puncture(CLP)procedure.The control group only flip the cecum without ligation,besides prevention group were injected with Lentinan for four weeks,other groups were injected with water before CLP.Lentinan treatment group were injected with Lentinan after CLP for four weeks.Endotoxin,blood gas analysis and tumor necrosis factorα(TNF-α)of all groups were detected.Results:Endotoxin levels of treatment group and prevention group significantly lower than those in the model group(P〈0.05)),PaO2 of experimental group was lower and PaCO2 was higher than those in the control group(P〈0.05),PaO2 of prevention group was higher and PaCO_2 was lower than those in model group(P〈0.05).TNF-αcontent of model group,treatment group were higher than control group(P〈0.05);Prevention,treatment group were lower than model group(P〈0.05);Prevention and treatment group were higher than the control group(P〈0.05).Conclusion:Lentinan can significantly protect the sepsis and decrease the content of TNF-αserum,which is the important mechanism of protecting the sepsis impossibly.
