基质金属蛋白酶-9对兔室壁瘤形成影响的实验研究

Effects of matrix metalloproteinase-9 on formation of left ventricular aneurysm in rabbits

目的:观察室壁瘤形成基质金属蛋白酶-9(MMP-9)血清浓度与心肌阳性细胞表达特点,探讨其对室壁瘤形成的作用。方法:20只新西兰白兔结扎冠状动脉制作室壁瘤模型,于术后4周行超声心动图检查并抽血,采用酶联免疫吸附法测定MMP-9的血清浓度,使用抗生物素-生物素-过氧化物酶复合法测定MMP-9在心肌组织中阳性表达细胞数。使用常规超声心动图判断实验兔有无室壁瘤形成,有室壁瘤形成纳入室壁瘤组,无室壁瘤形成纳入心肌梗死组。正常健康新西兰兔6只作为对照组。结果:术后4周室壁瘤组MMP-9血清浓度及阳性表达细胞数明显高于心肌梗死组与正常对照组,差异有统计学意义(P<0.05),与对照组比较,心肌梗死组MMP-9血清浓度及阳性表达细胞数变化不明显,差异无统计学意义(P>0.05)。结论:MMP-9对室壁瘤的形成有一定的促进作用。

Objective：To study the effect of matrix metalloproteinase-9（MMP-9）on the formation of left ventricular aneurysm（LVA）.Methods：Rabbit models of LVA were established in 20 New Zealand rabbits.Echocardiography were used to diagnose the formation of LVA at postoperative 4 weeks,and the MMP-9serum concentration was measured by the ELISA method.The double antibody sandwich ABC method was used to detect the expression of MMP-9in myocardial tissue.The animals which formed ventricular aneurysm were enrolled in the LVA group,which no formed ventricular aneurysm were enrolled in acute myocardial infarction group.Six New Zealand rabbits as the control group.Results：There were significant increase in the serum concentration and the positive expression cell of MMP-9in the LVA group than that in the control group and the myocardial infarction group,the differences were statistical significance（P〈0.05）.Compared with the control group,there was no obvious change in the serum concentration and the expression positive cell of MMP-9in the acute myocardial infarction group at postoperative 4 w,the differences were not statistically significant between the control group and the acute myocardial infarction group（P〈0.05）.Conclusion：MMP-9maybe promote an effect on the formation of LVA to a certain extent.
