摘要
目的:研究低氧训练状态下心肌组织细胞凋亡和血管新生的情况。方法:将60只健康雄性SD大鼠,随机分为6组,运动组采用负荷跑台训练8w,一周训练6d,运动量从最初的10 m/min、时间为15min增加到第8周的速度为25 m/min、时间为50 min,每周逢双在海拔1500 m的低氧环境下进行低氧训练,低氧浓度从最初的1500 m海拔高度增加到训练结束的3600 m海拔高度。结束训练后,采用戊巴比妥钠麻醉后取材。采用HE染色、TUNEL法进行细胞凋亡检测,Bcl-2、Bax和CD34的表达采用蛋白免疫组织化学方法进行检测,毛细血管的生成情况使用显微图象分析。结果:1)低氧对照组心肌组织细胞开始出现凋亡,血管生成没有;2)运动组心肌组织细胞有凋亡发生,血管生成也开始出现;3)低氧训练组与其它组别在凋亡方面没有显著性差异(p≥0.05),但在血管新生方面表现出很大的差异性(p≤0.05)。以上情况提示低氧复合运动不会加重心肌组织细胞凋亡,但会促进血管新生。
Objective:To study the effects of Hypoxic Training to Myocardial Apoptosis and Angiogenesis in Rats.Methods:60 male SD rats were randomly divided into six groups.8 weeks progressive treadmill exercise program were designed with intensity starts from10m/min for 15 min at the first week to 26m/min for 50 min in the last week and hypoxia degree from 1500 m at the first week to 3600 m in the last week.The protein expression of CD34,Bcl-2,Bax in the cardiomyocytes were analysis with HE staining,TUNEL,and immunity histochemistry.Microscopic image analysis capillary density,optical density level,express area.Results:1) when simple hypoxic myocardial tissue apoptosis is exsited,but angiogenesis is not happened.2) the move group reduced myocardial tissue Bcl-2expression,Bax expression change is not obvious,the Bcl-2/Bax ratio decreased,promote apoptosis,expression of CD34 positive things began to emerge.3) apoptosis in hypoxic exercise group compared with the low oxygen groups,sports group difference is not very big,but show great differences in terms of angiogenesis.Prompt low oxygen compound movement doesn't increase the apoptosis of myocardial cells,but will promote new blood vessels.
