贵州不同产地钩藤的总生物碱含量测定 被引量:11

摘要 为贵州钩藤总生物碱含量的测定及质量标准制定提供参考,以钩藤碱为对照品,采用酸性染料比色法在pH 3.6的邻苯二甲酸氢钾-盐酸缓冲液条件下,于波长413nm处测定总生物碱的含量。结果表明:钩藤碱含量(x)在0.022 4~0.358 4mg与吸光度(y)有良好的线性关系,其回归方程y=0.500 1x-0.043 4,R^2=0.99 9;平均回收率为97.11%,RSD为1.93%,该方法灵敏度高,准确度好,重现性好。贵州不同来源钩藤的总生物碱平均含量存在差异,即贵州黎平无柄果钩藤(2.218 3 mg/g)>贵州开阳钩藤(1.834 9mg/g)>贵州剑河无柄果钩藤(1.456 9mg/g)>贵州黎平钩藤(1.089 5mg/g)。 To provide references for determination of total alkaloids in U. rhynchophylIa and quality standard formulation in Guizhou. Taking rhynchophylline as the standard reference, the total alkaloids were tested at 413 nm with acid dye colorimetry in potassium acid phthalate-hydrochloric acid buffer solution(pH 3. 6). Results: The rhynchophylline content(x) was in good linearity with absorbance(y) over the range of 0. 022 4--0. 358 4 mg. The regression equation was y=0. 5001x--0. 043 4,R^2=0. 999, and the average recovery rate was 97.11%, with RSD 1.93(n=6). The method is of high sensitivity and accuracy, and excellent reproducibility. The average content of total alkaloids in U. rhynchophylla from different producing areas in Guizhou were different, following the sequence of Uncaria sessili fructus from Liping (2. 218 3 mg/g)〉U, rhynchophylla from Kaiyang(1. 834 9 mg/g)〉U, sessili fructus from Jianhe (1. 456 9 mg/g)〉U, rhynchophylla from Liping(1. 0895 mg/g).
