血清蛋白质组学技术筛选寻常型银屑病不同中医证型的差异蛋白 被引量:1

Study on Selecting Psoriasis Vulgaris Serum Proteins in Different TCM Syndrome Types Using Proteomics Technologies

摘要 目的:研究不同证型寻常型银屑病患者血清中的差异表达蛋白,从蛋白质组学探讨寻常型银屑病的发病机理及微观辨证物质基础。方法:收集寻常型银屑病患者血瘀证、血虚证、血热证各10例,健康人10例的血清。应用蛋白提取、定量、双向电泳技术、质谱鉴定和数据库检索,找出寻常型银屑病组与正常组、寻常型银屑病组不同证型之间的差异蛋白。结果:寻常型银屑病组与正常组间差异蛋白有20种,寻常型银屑病组患者血液中纤维蛋白原β链、血液结合素、纤维蛋白γ链、HPX蛋白、α-胰蛋白酶、载脂蛋白A-Ⅳ、补体C3、凝聚素(载脂蛋白,片段)、血清转铁蛋白、间-α-胰蛋白酶抑制剂H4重链(片段)、载脂蛋白E、补体C4-B、甘露糖结合蛋白-C、富亮氨酸-α-2-糖蛋白及血清白蛋白的表达明显升高,而血清白蛋白(片段)、甲状腺素转运蛋白、IgαC链、载脂蛋白L1、结合珠蛋白的表达则显著降低。其中,寻常型银屑病组不同证型之间也存在一定差异,补体C3、凝聚素、血清转铁蛋白等在血虚型寻常型银屑病患者的血清中表达高于其他两种证型的银屑病;纤维蛋白原β链、血液结合素等的表达在血热型寻常型银屑病患者血清中的表达水平高于血瘀型寻常型银屑病患者。结论:寻常型银屑病组和正常组以及寻常型银屑病不同中医证型间出现的血清蛋白差异表达,可能与寻常型银屑病的发病机制和银屑病不同证型的物质基础有关。 Objective: To study on the different syndromes of psoriasis vulgans of differentially serum expressed proteins in patients, to explore the pathogenesis of psoriasis vulgaris mechanism and microcosmic differentiation basis. Methods: The serum of psoriasis vulgaris patients with blood stasis syndrome and blood deficiency, blood-heat syndrome and healthy people were collected. Application of two-dimensional electrophnresis for protein extraction, quantitative, technology, mass spectrometry and database searches, to identify the different expression of serum prntein in psoriasis vulgaris group and normal group. Results: There are 20 species differential protein between psoriasis vulgaris group and normal group, the Fibrinogen beta chain, blood combined pigment, fiber protein gamma chain, HPX protein, Alpha-trypsin, contains lipoprotein a,-Ⅳ, and fill body C3, condensed pigment (contains fat protein, fragment), serum turned iron prolein, between-alpha-trypsin inhibitors H4 heavy chain (fragment), contains lipoprotein E, fill body C4-B, dew sugar combined protein-C, Leucine-rich alpha-2- protein and serum albumin expression in psoriasis vulgaris group significantly increased, while thyroid hormone transport proteins, IgαCchain, apolipoprotein L1, haptoglobin expression decreased. There were some differences between different syndrome types, fill body C3, condensed pigment, and serum turned iron protein in blood deticiency group was higher than the other two types. And the expression of fibrinogen beta chain and blood com- bined pigment in blood heat group was higher than the other two groups. Conclusion: The different expression of serum protein in psoriasis vulgaris group and normal group, and among different TCM syndromes, may be linked to the pathogenesis of psoriasis vulgaris psoriasis and different types of material.
