摘要
目的观察寿胎丸对控制性促排卵大鼠胚胎着床及雌激素、孕激素的影响。方法采用孕马血清促性腺激素(pregnant mare serum gonadotropin,PMSG)联合人绒毛膜促性腺激素(human chorionic gonadotropin,HCG)造成胚胎着床障碍模型,造模后将妊娠大鼠随机分为模型组、寿胎丸低剂量组、寿胎丸中剂量组、寿胎丸高剂量组和黄体酮组,正常组为不给药而自然合笼后妊娠者,每组10只,均于妊娠第1天开始治疗,寿胎丸低、中、高剂量组分别给予低、中、高剂量寿胎丸煎液灌胃,黄体酮组肌肉注射临床等效剂量黄体酮4 mg/(kg·d),正常组和模型组则每天等体积蒸馏水灌胃,连续治疗7 d,于妊娠第8天处死大鼠,留取血清和子宫组织,并观察比较6组大鼠胚泡着床点数。采用酶联免疫吸附测定法(enzyme-linked immune sorbent assay,ELISA)检测6组大鼠血清雌二醇(estradiol,E_2)、孕酮(progesterone,P)含量,Western blot法检测胚泡着床处子宫内膜雌激素受体(estrogen receptor,ER)、孕激素受体(progesterone receptor,PR)蛋白水平表达。结果与正常组比较,模型组的胚泡着床点数、E_2、P、ER及PR表达均降低,组间比较差异具有统计学意义(P<0.05);与模型组相比,寿胎丸中、高剂量组及黄体酮组大鼠胚泡着床点数、E_2、P、ER、PR表达均提高,组间比较差异具有统计学意义(P<0.05),而寿胎丸低剂量组E_2、ER表达虽提高,但与模型组相比差异无统计学意义(P>0.05);与黄体酮组比较,寿胎丸各剂量组胚泡着床点数、E_2、P水平均无统计学意义(P>0.05)。结论中、高剂量寿胎丸能提高促排卵大鼠的胚泡着床点数,其机制可能是提高雌激素、孕激素及其受体的水平,协调子宫内膜的容受性。
Objective To evaluate the mechanism of stimulating ovulation control rats by studying the effects of Shoutai pills on expression of estrogen,hormone and embryo implantation.Methods Pregnant mare serum gonadotropin(PMSG)combined with human chorionic gonadotropin(HCG)as the model rats of stimulating ovulation control.The rats with stimulate ovulation control were randomly divided into model group and low-,middle-and high-dose groups of Shoutai pills respectively given by gavage low-,middle-and high-dose of Shoutai pills,normal pregnancy group without any intervention,progesterone group intramuscular injected equivalent dose of progesterone[4 mg/(kg·d)](n=10/group),while model group and normal pregnancy group were given with equal volume distilled water.The rats were killed on the eighth day after continuous treatment for 7 days and embryo implantation was counted,blood was tested to detect the expressions of estradiol E2 and progesterone(P)with enzyme-linked immune sorbent assay(ELISA),and their decidual and placental tissues were co-cultured to detect the expressions of estrogen receptor(ER),progesterone receptor(PR) with Western Blot. Results The embryo implantation rate,E2,P,ER and PR of the model group were significantly lowerer than the normal pregnancy group(P〈0.05).Compared with model group,the embryo implantation rate,expressions of E2,P,ER and PR were increased in the middle-, high-dose groups of Shoutai pills and progestin group(P〈0.05),while it was not significantly difference in E2 and ER expressions between the low-dose group of Shoutai pills and model group(P〉0.05).The embryo implantation rate,expressions of E2 and P were not obviously difference between all groups of Shoutai pills and progestin groupn(P〉0.05).Conclusion The mechanism about middle-and high-dose groups of Shoutai pills can improve the expressions of E2,P,ER and PR,which may promote embryo implantation and endometrial receptivity.
