摘要
目的建立检测人血浆中布洛芬浓度的高效液相色谱法。方法以Agilent TC-C_(18)反相柱(150mm×4.6mm,5μm)为色谱柱,流动相为30mmol·L-1醋酸铵-甲醇-乙腈(25∶55∶20 V/V/V);流速:1.0mL·min^(-1);检测柱温:25℃;检测波长:230nm。以乙酸乙酯为提取剂。结果布洛芬的低、中、高(2.0,8.0,32.0μg·mL^(-1))3种浓度平均相对回收率分别为101.5%,99.58%,98.76%,提取回收率分别为70.82%,72.21%,75.86%;日内、日间相对标准偏差(RSD)均低于15%(n=5);分析方法的检测限0.5μg·mL^(-1);线性范围为1.0~48.0μg·mL^(-1)。线性方程:Y=2.692X-1.26,r=0.9995(n=7)。结论该方法简单、快速、灵敏、准确,可用于布洛芬临床血药浓度监测和药动学研究。
OBJECTIVE To establish a method for determining the concentration of ibuprofenc in human plasma by HPLC.METHODS The drug from plasma was analyzed in a reverse phase HPLC system C_(18)column(150 mm×4.6mm,5μm);mobile phase consisted of 30mmol·L(-1) ammonioum-methanol-acetonitrile(25∶55∶20 v/v/v);the flow rate was 1.0mL·min(-1);the detection temperature was at 25℃,the detection wavelength was at 230 nm.Ethyl acetate was used as extracting solvent.RESULTS The average recoveries of ibuprofenc in low,middle and high concentrations(2.0,8.0,32.0μg·mL-1) were 101.5%,99.58% and 98.76%,respectively.The extraction recovery were 70.82%,72.21% and 75.86%,respectively.The intra-day and inter-day variation(RSD) was less than 15%(n=5).The calibration curve of ibuprofenc showed good linearity,r=0.9995(n=7),over the range of 1.0-48.0μg·mL(-1).The minimum detectable concentration of ibuprofenc was 0.5μg·mL(-1).The regression equation was Y=2.692X-1.26.CONCLUSION The method is sensitive,quick,simple and accurate,it can be used for clinical drug monitoring and pharmacokinetics studies of ibuprofenc.
