外源性硫化氢对大鼠睾丸缺血再灌注损伤的保护作用 被引量:2

摘要 目的探讨外源性硫化氢对大鼠睾丸缺血再灌注损伤的保护作用。方法选取成年雄性Wistar大鼠30只随机分为3组,每组各10只:对照组(A组)、睾丸缺血再灌注损伤组(B组)、睾丸缺血再灌注损伤+硫化氢干预组(C组)。B组和C组大鼠建立睾丸扭转复位模型,C组在再灌注前腹腔注射外源性硫化氢(100μmol/kg)。HE染色观察睾丸组织病理改变;测定丙二醛(MDA)、超氧化物歧化酶(SOD)含量;采用原位缺口末端标记检测细胞凋亡指数;免疫组化检测睾丸组织中的Bax,Bcl-2和Caspase-3的表达情况。结果 B组可见生精上皮排列紊乱,生精细胞广泛脱落。与A组比较,B组中MDA含量和生精细胞凋亡指数明显升高,SOD活性明显下降,差异有统计学意义(P<0.05),B组中Bax/Bcl-2比值及Caspase-3表达明显高于A组;C组明显改善睾丸缺血再灌注损伤引起的变化,C组中MDA含量、SOD活性、生精细胞凋亡指数与B组比较,差异有统计学意义(P<0.05)。结论外源性硫化氢可减轻氧化应激水平和抑制生精细胞凋亡,从而在睾丸组织在缺血再灌注损伤过程中发挥保护作用。 Objective To investigate the protective effects of hydrogen sulfide (H2S) on the testicular ischemia-reperfu- sion injury in rats. Methods Thirty adult male Wistar rats were randomly divided into 3 groups, with 10 rats in each group, control group (group A), testicular ischemia-reperfusion injury group (group B), testicular ischemia reperfusion injury + H2S intervention group (group C). The rats of group B and C underwent testicular torsion lasting 2 hours, H2S (100 μmol/kg) was injected intraperitoneally before reperfusion in group C. HE staining was used to observe the patho- logical changes of testis; Malondialdehyde (MDA) and superoxide dismutase (SOD) content were determined; cell apoptosis index was detected by dUTP nick end labeling; the expression of Bax, Bcl-2 and Caspase-3 in testis tissue were detected by immunohistochemistry. Results After ischemia-reperfusion injury, the left testicular tissue showed desqua- mation of epithelial cells in the lumen, disordered arrangement of spermatogenic cells. Compared with the group A, MDA content and the spermatogenic cell apoptosis index were significantly increased, and SOD activity significantly decreased in group B, the differences were statistically significant (P 〈 0.05). The expression of Bax/Bcl-2 ratio and Caspase-3 in group B were significantly higher than those in group A; the group C obviously improved the changes of testicular IRI, MDA content, SOD activity, the apoptosis index in group C were compared with group B, the differences were statis!ically significant (P 〈 0.05). Conclusion H2S reduces the level of oxidative stress and inhibits the apoptosis of spermatogenic cells, it plays a protective role in the process of ischemia-reperfusion injury.
