摘要
以橡胶草根尖和嫩叶为材料,对其染色体制片技术的取样时间、预处理试剂、解离方法及解离时间进行比较研究。结果表明:橡胶草根尖和嫩叶取样最佳时间均为上午8:00~10:00;2 mmol/L 8-羟基喹啉溶液室温预处理4 h效果最好;根尖和嫩叶用1 mol/L盐酸于60℃下分别酸解10和12 min效果良好。
The root tips and young leaves of Taraxacum kok-saghyz Rodin were used for chromosome squashing to compare the differences in chromosome squashing through sampling time, pretreatment chemical solution, hydrolysing methods and time of chromosome squashing.The results showed that the root tips and young leaves were best sampled at about 8:00 ~10:00 am, pretreated in 2 mmol/L8-hydroxyquinoline solution at a room temperature for 4 hours and hydrolysed with 1 mol/L HCl at 60℃for 10 and 12 min respectively for chromosome squashing.
