摘要
目的快速、准确地鉴定口岸截获的5只蜚蠊。方法对中山港口岸截获的5只蜚蠊,分别进行形态和DNA条形码鉴定,比对5只样本间的DNA条形码序列,并将DNA条形码序列提交国际公共数据库,描述截获样本的形态特征,为今后口岸鉴定蜚蠊提供参考。结果经形态鉴定,截获的雄性蜚蠊为斯科特卡蠊Cartoblatta scorteccii,属于蜚蠊目,蜚蠊总科,蜚蠊科,卡蠊属。截获的雌雄个体间的DNA条形码序列差异均小于1%,判断这5个体为同一种类。经科技查新,确认为中国未见分布种,且为国内口岸首次截获。结论应用形态鉴定和DNA条形码相结合的鉴定方法可以有效提高外来物种的种类鉴别能力和准确度。
Objective To identify five intercepted cockroach rapidly and accurately. Methods To identify these five individuals intercepted at Zhongshan port with morphology and DNA barcoding techniques,compared the sequences among these five cockroaches,and submitted the DNA barcodes to the public data system. The morphological characters of the intercepted samples were illustrated as well,both the DNA barcodes and morphological characters would provide references for further identification of this species in the future. Results The male individual was identified as Cartoblatta scorteccii,belonging to Blattidae,Blattoidea,Blattodea. They were identified as the same species to the male based on that the inter-individual's DNA barcodes differences were all less than 1%. C. scorteccii is a non-recorded species in China and it is intercepted for the first time at Chinese ports based on Sci- Tech novelty search. Conclusion DNA barcoding is a strong supplementary tool for morphology taxonomy. Combining morphology and DNA bareoding techniques together could provide a good tool for the identification of intercepted species rapidly and accurately.
