FGF21对H9C2心肌细胞H/R损伤及Angpt2、GLUT1表达的影响

摘要 目的探讨FGF21对H9C2心肌细胞缺氧/复氧(H/R)损伤及Angpt2、GLUT1表达的影响。方法体外培养H9C2心肌细胞并分为H9C2组、H/R组、H/R+FGF21组,H9C2组即H9C2细胞阴性对照组,H/R组给予缺氧6 h/复氧24 h处理,H/R+FGF21组则在细胞复氧时给予FGF21(1μg/m L)处理。利用FCM检测各组细胞凋亡率,QPCR及Western blot检测各组Angpt2、GLUT1、Caspase-3的表达水平。结果 H/R组细胞凋亡率显著高于H9C2组(P<0.01)。与H/R组比较,H/R+FGF21组细胞凋亡率显著降低(P<0.01)。QPCR及Western blot检测发现,与H9C2组比较,H/R组Angpt2、Caspase-3表达显著增加,GLUT1表达显著降低(P<0.01);与H/R组比较,H/R+FGF21组Angpt2、Caspase-3表达显著降低(P<0.05或P<0.01),GLUT1表达显著增加(P<0.05)。结论外源性FGF21可显著减轻H/R损伤所致H9C2细胞凋亡,抑制炎症因子Angpt2表达和增加GLUT1表达可能是其内在机制。 Objective To investigate the effect of FGF21 on H/R injury and the expression of Angpt2 and GLUT1 in H9C2 cells. Methods H9C2 cells were cultured in vitro and were randomly divided into H9C2 group, H/R group and H/R+FGF21 group. H9C2 group was negative control group, H/R group underwent H/R injury (6 h hypoxia followed by 24 h reoxygenation), H/R+FGF21 group received FGF21 (1 μg/mL) treatment at the beginning of reoxygenation. The apop- tosis rate of H9C2 cell was determined by FCM. The expression level of Angpt2, GLUT1 and Caspase-3 were detected by QPCR and Western blot. Results The apoptosis rate was significantly increased in H/R group than that in H9C2 group (P 〈 0.01). Compared with H/R group, the apoptosis rate of H/R+FGF21 group was significantly decreased (P 〈 0.01). Detection of QPCR and Western blot showed, compared with H9C2 group, the expression of Angpt2 and Caspase-3 in H/R group were increased significantly, GLUT1 expression was decreased significantly (P 〈 0.01). Compared with H/R group, the expression of Angpt2 and Caspase-3 in H/R+FGF21 group were significantly decreased (P 〈 0.05 or P 〈 0.01), GLUT1 expression was increased significantly (P 〈 0.05). Conclusion FGF21 can significantly reduce the H9C2 cell apoptosis induced by H/R injury, inhibiting Angpt2 expression and increasing GLUT1 expression may be the inner mechanisms.
