上皮性卵巢癌中CXCR4与MMP-9的表达及其相关性研究 被引量:7

摘要 目的:探讨趋化因子受体4(CXCR4)和基质金属蛋白酶-9(MMP-9)在上皮性卵巢癌中的表达及其相关性。方法:选择30例上皮性卵巢癌(浆液性卵巢癌、黏液性卵巢癌)患者组织标本作为恶性组,同期14例卵巢良性上皮性肿瘤(浆液性囊腺瘤、黏液性囊腺瘤)患者组织标本作为良性对照组,采用免疫组化法检测两组组织中CXCR4及MMP-9蛋白的表达,以及与临床病理特征的关系,并分析恶性组中CXCR4、MMP-9蛋白表达的相关性。结果:CXCR4、MMP-9主要表达于细胞质中,呈棕黄色;恶性组CXCR4、MMP-9蛋白表达阳性率高于良性对照组,差异有统计学意义(P<0.05);Spearman相关性检验显示,CXCR4和MMP-9的表达与淋巴结转移、病理分期、大网膜转移、腹水中找到癌细胞、术前CA125值>600有关(P<0.05);配对设计关联性分析显示,CXCR4与MMP-9表达呈正相关(r=0.356,P<0.05)。结论:CXCR4、MMP-9在上皮性卵巢癌的侵袭转移过程中起重要的作用,两者具有协同关系。 Objective:To study the expression and correlation of chemokine receptor 4( CXCR4) and matrix metalloproteinase 9( MMP-9) in epithelial ovarian carcinoma. Methods:Thirty histological specimens of epithelial ovarian carcinoma( serous ovarian cancer and mucous ovarian cancer) were chosen as malignant group,and 14 specimens of benign ovarian epithelial tumor( serous ovarian adenoma and mucous ovarian adenoma) in the same time as benign control group. CXCR4 and MMP-9protein expression were determined with immunohistochemical method,and their correlation in malignant tissue was analyzed. Results:CXCR4 and MMP-9 were claybank and mainly expressed in cytoplasma. The positive expression rates of CXCR4 and MMP-9 in malignant group were significantly higher than in benign control group( P〈0. 05); Spearman correlation test results showed that the expressions of CXCR4 and MMP-9 were correlated with lymphatic metastasis,pathological stages,omentum metastasis,positive cancer cell detection rate in ascitic fluid and big CA125 value(〉 600) before operation( P〈0. 05); Paired design relevance analysis showed there was a positive correlation between expression of CXCR4 and MMP-9 in epithelial ovarian carcinoma( r = 0. 356,P〈0. 05). Conclusions:CXCR4 and MMP-9 both play an important role in the invasion and metastasis of epithelial ovarian cancer,and have cooperative relationship to each other.
