摘要 目的:建立天然丹皮酚与合成丹皮酚的鉴别方法。方法:以3种不同产地天然丹皮为原料,采用水蒸气蒸馏法制备天然丹皮酚。以间苯二酚为原料,分别经过乙酸乙酰化和硫酸二甲酯甲基化得到合成丹皮酚,以水蒸汽蒸馏法进行纯化。采用HPLC法、薄层色谱(TLC)、气相-质谱法(GC-MS)对天然丹皮酚与合成丹皮酚进行鉴别。结果:天然丹皮酚与合成丹皮酚的HPLC色谱图中,杂质的出峰时间有明显差别;在TLC谱图中合成丹皮酚有杂质斑点;在GC-MS谱图中合成丹皮酚有杂质峰(2,4-二甲氧基苯乙酮)。以上3种方法均能鉴别天然丹皮酚与合成丹皮酚。结论:本研究建立的方法简便、快速、高效,可用于鉴别天然丹皮酚与合成丹皮酚。 Objective: To establish a method for the identification of natural and synthetic paeonol. Methods: Natural pae- onol was extracted from Cortex Moutan Radicis produced in three different regions by steam distillation. Synthetic paeonol was synthesized from resorcinol through two steps of acetylation with acetic acid and methylation with dimethyl sulfate, and then was purified by steam distillation. HPLC, thin layer chromatography (TLC) and gas chromatography-mass spectrometry (GC-MS) were employed to identify the two kinds of paeonol. Results: Retention time of impurities was significantly different in the HPLC chromatograms of natural and synthetic paeonol. Impurity spots appeard in the TLC photograms of the synthetic pae onol sample. Impurity peak of 2,4-dimethoxy acetophenone could be detected only in the GC-MS chromatograms of synthetic paeonol. Therefore,the natural and synthetic paeonol could be identified by HPLC, TLC and GC-MS methods. Conclusion: The method established by this study is simple, rapid,efficient,and could be used for the identification of natural and synthetic paeonol.
