六味地黄方药饼对免疫抑制兔脾脏指数及免疫因子的影响

Effects of Liuwei Dihuang recipe cake on spleen index and immune factors in immunosuppressive rabbits

目的观察在以六味地黄汤方制成药饼上艾灸对免疫抑制兔脾脏指数及细胞因子干扰素(IFN)-γ、白细胞介素(IL)-2及IL-4的影响。方法将大耳白兔按随机数字表分为空白组、模型组、隔药饼灸组和假药饼灸组,每组10只,共40只。除空白组外,后3组动物环磷酰胺注射于腹腔,完成免疫抑制动物模型的制备。以六味地黄汤方和面粉分别制成中药药饼和假药饼,将药饼放置于神阙、关元等穴,艾柱置于药饼上,进行隔药饼灸。隔日施灸治疗,共计10次。治疗结束第2天,动物麻醉后取脾脏计算其脾脏指数,脾组织匀浆备测,酶联免疫吸附法(ELISA)测定上清液中IFN-γ、IL-2、IL-4含量。结果环磷酰胺连续注射造成动物免疫抑制模型后,脾脏指数显著升高,与空白组比较,差异具有统计学意义(P<0.01);隔药饼组和假药饼组脾脏指数均与模型组比较差异有统计学意义(P<0.01),与空白组比较差异无统计学意义(P>0.05)。在免疫因子含量的变化中,2治疗组IFN-γ含量,与空白组比较均降低,差异有统计学意义(P<0.05),与模型组比较差异有统计学意义(P<0.05),2治疗组之间差异无统计学意义(P>0.05)。IL-4含量中,模型组、假药饼组与空白组比较,降低明显,差异均具有统计学意义(P<0.05);隔药饼灸组IL-4显著升高,与模型组、假药饼组比较差异有统计学意义(P<0.05),与空白组比较差异无统计学意义(P>0.05)。结论艾灸对免疫抑制兔免疫抑制状态有明显的调节作用,六味地黄汤方药饼对脾组织中IL-4含量的升高作用具有显著的特异性。

Objective To determine the effects of Liuwei Dihuang decoction prepared cake-seperated moxibustion on the spleen index and the cytokines IFN-γ IL-2 and IL-4 in immunosuppressive rabbits. Methods A total of 40 white rabbits were randomly divided into the blank group, model group, cake-separated moxibustion group and sham cake-separated moxibustion group （n=10 each）. In addition to the blank group, the latter three groups of animals received intraperitoneal injection of cyclophosphamide to complete the preparation of immunosuppressive animal model. The traditional Chinese medicine cakes and sham cakes were prepared by Liuwei Dihuang decoction and flour, and were placed at the Shenque and Guanyuan acupoints, respectively. The moxa cone was placed on the cake for cake- separated moxibustion. The moxibustion treatment was performed every other day for a total of 10 times. At the second day after the treatment, the spleens of the rabbits were taken after anesthesia to calculate the spleen index. The spleen tissue homogenate was prepared for the test. The contents of IFN-T, IL-2 and IL-4 in the supernatant were determined by ELISA. Results The spleen index was significantly increased after continuous injection with cyclophosphamide for the immunosuppressive model, and there was statistically significant difference compared with that in the blank group （P〈0.01）. There was significant difference in the spleen index between the cake-separated moxibustion group and sham cake-separated moxibustion group （P〈0.01）, whereas there was no significant difference compared wih that in the blank group （P〉0.05）. In the content change of the immune factors, the content of IFN-γ in the two treatment groups was significantly decreased compared with that in the blank group, and there was significant difference between the two groups （P〈0.05）. There was significant difference in the content of IFN-γ between the treatment groups and the model group （P〈0.05）, whereas there was no significant difference between the two treatment groups （P〉0.05）. The IL-4 content was significantly decreased in the model group and the sham cake-separated moxibustion group compared with that in the blank group, and the difference was statistically significant （P〈0.05）. The IL-4 content was significantly increased in the cake-separatod,moxibustion group, and there was statistically significant difference compared with that in the model group and the sham cake-separated moxibustion group. There was no significant difference in the IL--4 content between the sham cake-separated moxibustion group and the blank group （P〉0.05）. Con- clusion Moxibustion shows significant regulation on the immunosuppressive state of immunosuppressive rabbits. Li- uwei Dihuang decoction cake shows significant specificity on the increase of IL-4 content in spleen tissues.
