人五聚素3可溶性CD40配体及核因子-κB与脑梗死的相关性研究

Correlation between PTX3, sCD40L and NF-κB and cerebral infarction

目的研究人外周血人五聚素3(PTX3)、可溶性CD40配体(sCD40L)及核因子-κB(NF-κB)与脑梗死的相关性,并进一步探讨此3种炎性因子之间的关联及可能的信号通路。方法选取急性脑梗死患者84例作为病例组,及同时期体检中心健康体检者80名作为对照组。分别采用酶联免疫吸附测定法(ELISA)和流式细胞术(FCM)检测血浆sCD40L、PTX3的浓度,及人外周血单个核细胞(PBMC)中NF-κB的阳性表达率。结果病例组PTX3及sCD40L浓度、NF-κB阳性率均显著高于对照组(P<0.05)。病例组NF-κB与PTX3表达水平之间存在正相关(r=0.252,P=0.021),sCD40L与PTX3血浆水平之间存在正相关(r=0.342,P=0.001)。结论 NF-κB、sCD40L及PTX3均是与脑梗死密切相关的炎性因子,三者之间可能是相关炎症信号通路中的上下游炎症因子。

Objective To determine the correlation between human peripheral blood pentosan 3 （PTX3）, soluble CD40 ligand （sCD40L）, nuclear factor-κB （NF-κB） and cerebral infarction, and to further investigate the correlation between these three inflammatory factors and the possible signaling pathways. Methods Eighty-four patients with acute cerebral infarction were included as the case group and a contemporary cohort of 80 healthy subjects examined in the physical examination center were included as the control group. The concentration of plasma sCD40L and PTX3, and the positive expression rate of NF-κB in human peripheral blood mononuclear ceils （PBMC） were measured by enzyme-linked immunosorbent assay （ELISA） and flow cytometry （FCM）, respectively. Results The concentration of PTX3 and sCD40L and the positive rate of NF-KB in the case group were significantly higher than those in the control group （P〈0.05）. There was a positive correlation between the expression levels of NF-KB and PTX3 in the case group （r=-0.252, P=0.021）, and there was a positive correlation between the sCD40L and PTX3 plasma levels （r=0.342, P=0.001）. Conclusion NF-KB, sCD40L and PTX3 are inflammatory factors, which are closely related to cerebral infarction, and may be the upstream and downstream inflammatory factors in the relevant inflammatory signaling pathways.
