摘要
目的:比较不同方法处理的天龙水提物对乳腺癌MCF-7细胞株的抑制作用。方法:使用酶解法和甲基噻唑蓝(MTT)法,以蛋白质水解度、对乳腺癌细胞MCF-7的抑制率为指标,评价混合酶对发酵前后天龙粉的水解效果,以天龙粉水提物作为对照组,比较天龙粉酵解物和发酵前、后分别经胰蛋白酶和弹性蛋白酶混合酶解水提物对乳腺癌细胞的抑制作用。结果:混合酶对天龙粉的水解度(52.46%)大于对发酵天龙粉的水解度(10.71%);天龙粉混合酶解水提物对乳腺癌细胞的抑制率为正值,随浓度成正相关,天龙粉与发酵天龙粉酶前、酶后水提物对乳腺癌细胞的抑制率为负值。结论 :天龙粉混合酶解水提物对乳腺癌MCF-7细胞起到抑制作用,天龙粉与发酵天龙粉酶前、酶后水提物对乳腺癌细胞无抑制作用。
Objective To compare the different treatment methods of Tianlong water extract of inhibition effect on human breast cancer cell line MCF-7: Methods: Using enzyme hydrolysis method and methyl thiazolyl blue(MTT) method,the degree of protein hydrolysis and the inhibition rate on MCF-7 breast cancer cells as evaluating indicator,Evaluating the hydrolysis effect of mixed enzyme on the Tianlong powder before and after fermentation,Tianlong water extract as contrast group,comparing the inhibitory effect of water extract of Tianlong glycolysis and fermentation before and after are enzymatic hydrolysised by trypsin and elastase respectively on human breast cancer cells. Results: The hydrolysis degree(52. 46%) of Tianlong on mixed enzyme hydrolysis is higher than the hydrolysis degree(10. 71%) of fermentation Tianlong on it; Tianlong mixed enzymolysis water extract's inhibition rate on breast cancer cells is positive,with the positive correlation of concentration,Tianlong and fermentation Tianlong is enzymed before and after have negative inhibiting rate on breast cancer cells. Conclusion: Tianlong mixed enzymolysis water extract have inhibition on breast cancer cell MCF-7,Tianlong and fermentation Tianlong is enzymed before and after haven't inhibitory effect on breast cancer cells.
