摘要
为有效开展大花萱草的育种工作,采用TTC染色法对‘东方不败’、‘红运’、‘金娃娃’、‘吉星’、‘莎蔓’和‘红色海盗’6个大花萱草品种的花粉生活力进行了测定,并研究了在离体培养条件下,白砂糖和硼酸对其花粉萌发的影响。结果表明:在TTC染色试验中,6种大花萱草花粉的生活力是‘金娃娃’>‘红运’>‘东方不败’>‘红色海盗’>‘莎蔓’>‘吉星’。这6个品种花粉的最强活力在1天中出现的时间不同,其中‘东方不败’、‘红运’和‘红色海盗’花粉生活力的峰值出现在10:30时左右,‘吉星’和‘莎蔓’花粉生活力的峰值在9:30时左右,‘金娃娃’花粉生活力的峰值在8:30时左右。在离体萌发试验中,‘东方不败’、‘金娃娃’、‘吉星’、‘莎蔓’和‘红色海盗’花粉最适宜的萌发培养基均为白砂糖100g/L+硼酸50mg/L,其萌发率分别为27.13%、28.81%、5.37%、19.00%和21.62%;而‘红运’花粉萌发的适宜培养基为白砂糖150g/L+硼酸50mg/L,其萌发率为15.19%。
In order to effective breeding of Hemerocallis hybrida,the pollen viability of 6 varieties( H.‘Peachy Pink’,H. ‘Batimore oriole’,H.‘Stella de Oro’,H. cv.‘Butter Curls’,H. ‘Siloam Amazing Grace’ and H.‘Crimson Pirate’) were tested by TTC staining method,and the effects of sugar and boric acid on their pollen germination were studied in vitro culture conditions. The results showed that the pollen viability order of 6 H. hybrida varieties from high to low was‘Stella de Oro’,‘Batimore oriole’,‘Peachy Pink’,‘Crimson Pirate’,‘Siloam Amazing Grace’,‘Butter Curls’. In a specific day,the pollen viability of 6 varieties dynamic change over time,and their most active time appeared in different period. At 10:30 am,the pollen viability of ‘Peachy Pink’,‘Batimore oriole’ and‘Crimson Pirate’ were the highest. At 9:30am,the pollen viability of ‘Butter Curls’ and ‘Siloam Amazing Grace’ were the highest. When it was 8:30am,pollen viability of ‘Stella de Oro’ was the highest. In vitro germination test,the most suitable medium for pollen germination of ‘Peachy Pink’,‘Stella de Oro’,‘Butter Curls’,‘Siloam Amazing Grace’ and ‘Crimson Pirate’ were all sugar 100g/L+boric acid 50mg/L,and their germination rates were 27. 13%,28. 81%,5. 37%,19. 00% and 21. 62%,respectively. The most suitable medium for pollen germination of ‘Batimore oriole’ was sugar 150g/L + boric acid 50mg/L,and its germination rates was 15. 19%.
