摘要
目的:对远志多糖进行纯化分离,观察其具有药用功效的单一多糖,为其药用功效进一步研究奠定基础。方法:采用水提醇沉法提取远志粗多糖,用酶法和Sevage法除蛋白,活性炭脱色,葡聚糖凝胶Sephadex G-100分离纯化,并对分离得到的多糖进行初步的结构分析。结果:分离纯化得到的远志单一多糖,紫外可见光260 nm、280 nm处有多糖无蛋白质和核酸的吸收峰,在红外光谱也有表现出多糖的特征官能团。结论:推测该多糖为β-吡喃糖,而非α-吡喃糖,并且不含蛋白质。
Objective:To study purification and isolation of polysaccharides from Polygala tenuifolia. Method:This experiment used water extracting and alcohol precipitation extraction of polysaccharides from Polygala tenuifolia,removing protein by enzymatic and Sevage,decoloration by activated carbon,and isolation and purification by Sephadex G-100,and prove that the extraction of polysaccharides from a single component. At the same time by using chromatography,UV spectroscopy,and infrared spectroscopy to a preliminary study on its structure. Results:It shows that the polysaccharide from Polygala tenuifolia without protein,infrared spectroscopy has shows some its own special features. Conclusion:Speculate that single polysaccharide is beta-tetrahydropyranyl sugar,rather not alpha-tetrahydropyranyl sugar and nonprotein-containing polysaccharides.
