摘要
目的:测定两头尖多糖中单糖的组成。方法:两头尖多糖采用2 mol/L三氟醋酸水解后通过1-苯基-3-甲基-5-吡唑啉酮(PMP)柱前衍生,用高效液相色谱法梯度洗脱测定;检测波长250 nm。结果:两头尖多糖由D-葡萄糖、D-核糖、L-鼠李糖、L-阿拉伯糖、L-岩藻糖、D-甘露糖、D-半乳糖、D-木糖)组成。结论:高效液相色谱法测两头尖多糖的单糖组成,操作方法简便,具有良好的重复性及稳定性。
Objective:To determine monosaccharide composition in polysaccharide of Anemone raddeana Regel by high performance liquid chromatography. Method:The polysaccharide were hydrolyzed into monosaccharides with 2 mol/L trifluoroaceticacid.The monosaccharide obtained with 1-phenyl-3-methyl-5-pyrazolone(PMP)were separated by reversephase HPLC using gradient elution process,and monitored by ultraviolet detector at 250 nm. Result:The results showed that Anemone raddeana Regel compose of D-glucose,D-ribose,L-rhamnose,L-arabinose,L-fucose,D-mannose,D-galactose,D-xylose. Conclusion:The method was simple,has a good repeatability and stability and could be used to determine the monosaccharide composition of Anemone raddeana Regel polysaccharide.
