厄贝沙坦对2型糖尿病大鼠肾组织BGP表达的影响 被引量:3

Effect of irbesartan on the expression of osteocalcin in renal tissue of type 2 diabetic rats

摘要 目的探讨糖尿病肾病大鼠胰岛及肾脏骨钙素(BGP)表达的情况及厄贝沙坦对胰岛及肾脏骨钙素表达的影响。方法选择36只SD雄性大鼠给予链脲佐菌素(30 mg/kg)造糖尿病大鼠模型,随机分成糖尿病对照组(DM组)、50 mg/kg厄贝沙坦组(I组),另选取同龄大鼠作为正常对照组(NC组)。造模后采用灌胃法给药8周;ELISA法检测24 h尿白蛋白排泄率;图像分析测定胰腺及肾小球BGP。结果药物干预组的24 h尿白蛋白排泄率显著降低,BGP在胰腺的表达明显增多,而在肾脏的表达明显减少。结论骨骼分泌的骨钙素在胰岛细胞及肾脏呈阳性,提示其可能参与糖代谢及肾脏损害。厄贝沙坦治疗后肾脏骨钙素水平下降,提示其可能减少骨钙素在肾脏的沉积,改善肾功能,但机制有待证实。 Objective To investigate the expression of osteocalcin in islet and kidney of diabetic nephropathy rats,and the effect of irbesartan on the expression of osteocalcin in islet and kidney. Methods Thirty-six male Sprague-Dawley rats was given single intraperitoneal of streptozotocin( 30 mg / kg) to induce diabetes. The diabetic rats were randomly divided into three groups,diabetic control group( group DM),50 mg / kg irbesartan group( group I),normal rats receiving equal volume of water served as the normal control group( group NC). All the rats were sacrificed after 8 weeks of gavage.ELISA method was used to detect the excretion rate of 24 h urinary albumin,and the image analysis was used to detect the BGP in pancreatic and glomeruli of kidney. Results In the drugs intervention group,the 24 h urine albumin excretion rate significantly reduced,and the level of BGP significantly increased in pancreatic and decreased in glomeruli of kidney.Conclusion Osteocalcin secreted by Skeleton is positive in the kidney and islet cells,suggests that it may be involved in glucose metabolism and kidney damage. After treated with irbesartan,the BGP level decreases in kidney,suggesting that it may reduce the osteocalcin deposition,improve kidney function,but the mechanism is yet to be confirmed.
