摘要
目的:研究稀土元素镧与丹参毛状根中活性物质及其生物合成中关键酶基因表达的量效关系,结合Hormesis效应理论探讨镧调控丹参次生代谢产物的阈值,以期为丹参中次生代谢产物调控及镧肥的开发提供依据。方法:采用HPLC法测定丹参毛状根活性成分含量,采用TIANGEN多糖多酚植物总RNA提取试剂盒提取RNA,采用Takara反转录试剂盒合成cDNA,采用PCR荧光定量试剂盒进行实时定量检测,采用SPSS软件进行数据分析。结果:镧处理对丹参活性成分积累及关键酶基因表达量影响显著,丹参酮Ⅰ、丹参酮ⅡA、迷迭香酸和丹酚酸B表现出低浓度促进高浓度抑制的"抛物线"趋势;AACT、C4H、4CL、TAT和HPPR五个关键酶基因表达量同样表现出低促高抑的"抛物线"趋势,该趋势符合Hormesis理论曲线。结论:根据Hormesis理论分析,镧对丹参的促进浓度阈值上限在0.1~1.0 mmol·L^(-1)之间。
Objective: The effect of La on the accumulation of active constituent and key enzymes expression of Salvia miltiorrhiza hairy root were studied and furthermore signaling molecules mediating the synthesis of secondary metabolism was also definited in order to provide references for the reveal of synthesis mechanism of active constituent of Salvia miltiorrhiza hairy root inducing by La. Methods: The content of active constituents were detected by HPLC. RNA was extracted with RNA prep Pure RNA purification kit(TIANGEN). Results: The dose-effect relationship between La and active constituent and key enzymes expression of Salvia miltiorrhiza hairy root showed a parabolic trend. The trend was in line with Hormesis effect curve. Conclusion: According to the Hormesis theory, the concentration upper threshold of La to promote Salvia growth is between 0. 1 - 1.0 mmol· L^-1.
