摘要
本研究利用乙醇初步提取发酵液中透明质酸,确定最佳提取方案为乙醇体积分数60%,洗涤次数为两次。然后,以透明质酸含量为指标,在单因素试验的基础上,采用根据Box-Behnken原理设计的响应面法优化透明质酸纯化工艺。方差分析和交互作用分析表明,最佳纯化工艺为NaCl浓度45 g/L,温度55℃,pH值9.6,在此工艺条件下,透明质酸含量为93.71%。本工艺所生产的透明质酸纯度较高,符合化妆品、医药行业标准,具有广泛的应用价值。
In this research,the hyaluronic acid was preliminarily extracted from fermentation broth using alcohol.The best extraction parameters were ethanol volume fraction of 60% and washing for two times.Using the content of hyaluronic acid as indicator,single-factor experiments were performed.Then the response surface analysis based on Box-Behnken experimental design was carried out to optimize the purification process of hyaluronic acid.Through analysis of variance and interaction,the optimal purification process was Na Cl additive amount of 45 g/L,temperature of 55℃ and p H value of 9.6.Under the optimized conditions,the actual content of hyaluronic acid reached up to 93.71%.By this process,the content of hyaluronic acid was higher,which conformed to the stands of cosmetic and pharmaceutical industries,so it had extensive application value.
