摘要
目的:研究1700008O03Rik(RIKEN cDNA 1700008O03)基因在B6小鼠睾丸中从出生到性成熟过程的表达特征,并初步探讨其参与调控小鼠精子发生的作用。方法:小鼠基因表达谱芯片筛选得到睾丸特异性基因1700008O03Rik,本研究采用反转录PCR、实时定量PCR、Western印迹、免疫组化及免疫荧光等实验方法,检测1700008O03Rik基因在小鼠睾丸发育中的表达特征,并用生物信息学方法作相关分析。结果:1700008O03Rik基因在小鼠睾丸中高表达,且呈鼠龄依赖性增加;免疫组化结果表明其主要表达于长形精子胞浆。生物信息学分析结果显示人和小鼠的1700008O03Rik蛋白序列高度同源,进化树分析结果表明从低等哺乳动物到高等哺乳动物1700008O03Rik蛋白均高度保守。结论:1700008O03Rik为小鼠睾丸高表达基因,且主要表达于长形精子胞浆,其可能参与调控小鼠精子的成熟过程。
Objective: To investigate the characteristics of the expression of the RIKEN cDNA 1700008003 (1700008003Rik) gene in the testis of the mouse from birth to sexual maturity and its potential role in regulating spermatogenesis. Methods: Using mouse gene expression profile microarray, we screened the testis-specific gene 1700008003Rik from the mouse. We studied the expres- sion characteristics of the gene in the development of the mouse testis by reverse transcription PCR, quantitative real-time PCR, West- ern-blot, immunohistochemistry and immunofluorescence, and analyzed the structure of the 1700008003Rik protein and its homology with other species using the bioinformatic software. Results: 1700008003Rik gene was highly expressed in the testis of the mouse, increasing in an age-dependent manner, and mainly in the endochylema of oblong spermatozoa. Bioinformatic analysis revealed a high homology of the 1700008003Rik protein between human and mice, and phylogenetic tree analysis showed it to be highly conserved in mammalian evolution. Conclusion : 1700008003Rik is a highly expressed gene in the mouse testis, mainly in the endochylema of ob- long spermatozoa, which may be involved in the regulation of spermatogenesis in mice.
