摘要
目的:探讨姜黄素对糖尿病大鼠的心脏保护作用及作用机制。方法:通过腹腔注射链脲佐菌素诱导大鼠2型糖尿病。观察姜黄素对大鼠大动脉血流动力学、心脏功能、血糖、血脂、磷脂酰肌醇3-激酶(PI3K)、蛋白激酶B(AKT)、磷酸化蛋白激酶B(p-AKT)、丙二醛(MDA)和一氧化氮(NO)表达,以及过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GPx)、超氧化物歧化酶(SOD)和一氧化氮合酶(NOS)活性的影响。结果:姜黄素处理对糖尿病大鼠AKT表达无明显影响,但可降低血糖、血脂、MDA表达以及血流动力学变化,增加心脏收缩和舒张功能,促进PI3K、p-AKT、NO的表达,提升CAT、GPX、SOD和NOS的活性。结论:姜黄素可通过改善代谢紊乱和血流动力学变化而对糖尿病大鼠心脏具有保护功效,其作用机制与激活PI3K/AKT信号通路而抑制氧化应激和促进NO生成相关。
Objective:To explore the protective effect and mechanism of Curcumin(Cur)on cardiac function in diabetic rats.Method:The model of diabetic rats were induced by intraperitoneal injection of streptozocin(STZ).The effect and potential mechanism of Cur on diabetic rats was evaluated by testing hemodynamics,cardinal function,the activation of CAT,SOD,GPX and NOS,blood glucose,blood lipid and the expression of PI3 K,AKT,p-AKT,MDA and NO.Result:Cur treatment had no influence on the AKT expression in diabetic rats,but could reduce the blood glucose,blood lipids,MDA expression and hemodynamic changes,simultaneously increase systolic and diastolic function,promote the expressions of PI3 K,p-AKT and NO,and enhance the activation ofCAT,GPX,SOD.Conclusion:Cur has protective effects on cardiac function in diabetic rats by improving systolic and diastolic function of heart,metabolism disorder and the changes of hemodynamics.The mechanism is associated with suppressing oxidative stress and promoting NO generation though inducing the activation of PI3K/AKT signal pathway.
