摘要 目的建立复方枣参片中斯皮诺素和总皂苷的含量测定方法。方法 HPLC法测定斯皮诺素采用Agilent Zorbax SB C_(18)柱(250mm×4.6 mm,5μm),柱温25℃,流动相为乙腈-水,采用梯度洗脱,流速为1.0 ml/min,检测波长为335nm。UV法测定总皂苷采用Amberlite XAD-2大孔树脂柱层析后,香草醛-冰醋酸法显色测定,检测波长为560nm。结果斯皮诺素和总皂苷的线性范围分别为20~100μg/ml(r=0.999 0)和40~200μg/ml(r=1.000 0),准确度平均值分别为99.55%和99.85%。结论本方法准确可靠,可作为复方枣参片的质量控制依据。 Objective To establish methods for the determination of spinosin and total saponins in compound Zaoshen tablet. Methods The separation of spinosin was performed on Agilent Zorbax SB Cls column (250 min× 4.6 mm, 5 /,m) at 30 ℃ with water and acetonitrile as mobile phase for gradient elution at the flow rate of 1.0 ml/min. The detection wavelength was 335 nm. Determination of total saponins was fulfilled by column chromatography followed by UV spectrometer with vanillin glacial acetic acid colorimetry. The detection wavelength was 560 nm. Results I.inear range of spinosin was 20-100 μg/ml (r=0. 999 0) and total saponins was 40-200 μg/ml (r= 1. 000 0). The average recoveries for accuracy tests were 99.55% and 99.85% respectively. Coneluslon Those methods are accurate and reliable. They can be used for the quality control of corn pound Zaoshen tablet.
