摘要
从地方耐高温品种中克隆出具有完全读码框的热激因子TaHSF3,编码315个氨基酸序列。系统进化树分析表明:TaHSF3属于热激因子(HSF)B2类,与大麦、水稻、玉米和大豆相似性较高;正常生长条件下TaHSF3基因在小麦穗部有高的表达量;在高温和冷冻胁迫处理下TaHSF3能高度表达。TaHSF3基因有利于应对极端温度,可以作为1个候选基因应用在小麦抗性分子育种中,将有可能培育出耐热的小麦新材料。
PCR method was used to clone TaHSF3 gene from heat-resisting varieties. The TaHSF3 had an open reading frame of 315 amino acids. Phylogenetic analysis showed that TaHSF3 belonged to Class B2 of HSF, sharing high similarity protein sequence to barley, rice, maize and soybean; the expression levels of TaHSF3 were highly up-regulated by heating and cold treatments and also high in spikes of wheat. Therefore, TaHSF3 might enhance the extreme temperature tolerance of wheat plants, and as a candidate gene to improve wheat heat tolerance through genetic engineering and might cultivate new strains of wheat.
