摘要
目的探讨Th22细胞及其细胞因子与原因不明复发性流产(URSA)发病的关系。方法采用流式细胞术检测32例URSA女性和20例正常早孕女性外周静脉血Th22细胞频数,采用ELISA法检测血清中IL-22和TNF-α细胞因子表达水平。结果 URSA组外周血Th22细胞的百分率高于正常对照组,差异有统计学意义(P<0.01)。URSA组外周血和蜕膜组织的IL-22和TNF-α表达水平明显高于正常对照组,差异有统计学意义(P<0.01)。URSA组患者的外周血和蜕膜组织中AHR mRNA表达明显高于正常对照组,差异有统计学意义(P<0.01)。URSA组患者与正常对照组患者外周血及蜕膜组织中IL-22细胞因子水平与Th22细胞百分比均呈正相关。URSA组患者与正常对照组患者外周血及蜕膜组织中Th22细胞百分比与AHR mRNA表达水平均呈正相关。结论 Th22细胞及其细胞因子IL-22的表达增高可能是导致URSA发病的重要原因。
Objective To explore the relationship of Th22 cells and their related cytokines with unexplained recurrent spontaneous abortion( URSA).Methods To detect the frequencies of Th22 cell in the peripheral vein blood of 32 URSA patients and20 cases of normal early pregnant women by flow cytometry,and to detect the cytokines levels of serum IL-22 and TNF-α by enzyme-linked immunosorbent assay( ELISA).Results The average percentage of Th22 cells in the peripheral blood and decidual of URSA group were significantly higher than that in the control group,and the differences were statistically significant( P〈0.01).The levels of cytokines IL-22 and TNF-α in the peripheral blood and decidual of URSA group were significantly higher than that of the control group,and the differences were statistically significant( P〈0.01).The mRNA expression in AHR of decidua and peripheral blood in URSA group were significantly higher than that of the control group,and the differences were statistically significant( P〈0.01).The correlation between the levels of cytokine IL-22 and the percentage of Th22 cells in peripheral blood and decidua of URSA group and control group were positive.The percentage of Th22 cells in the peripheral blood and decidua in URSA patients and control patients had positive correlation with the expression levels of mRNA in AHR.Conclusion Maybe Th22 cells and their related cytokine IL-22 are the main cause of URSA.
