8周爬梯负重训练诱导大鼠比目鱼肌和趾长伸肌PGC1α/FNDC5/MSTN差异表达 被引量:1

摘要 目的:观察8周爬梯负重训练对SD大鼠比目鱼肌和趾长伸肌PGC1α/FNDC5/MSTN表达及血清Irisin浓度的影响。方法:20只SD大鼠随机平均分为对照组(Con组,n=10)和爬梯训练组(Lad组,n=10),Lad组采用改进的爬梯负重抗阻力量训练模型,3天训练1轮,共进行8周。训练结束后48小时取比目鱼肌和趾长伸肌,用定量PCR和Western Blot法检测肌肉组织中PGC1α/FNDC5/MSTN的基因和蛋白表达,用Elisa法检测血清Irisin浓度。结果:与Con组相比,Lad组比目鱼肌PGC1α表达显著降低(P<0.05)、FNDC5表达有降低趋势,MSTN表达显著增加(P<0.05);而趾长伸肌PGC1α表达显著增加(P<0.05)、FNDC5表达有增加趋势,MSTN表达显著降低(P<0.05);血清Irisin浓度无明显差异(P>0.05)。结论:8周爬梯负重训练诱导SD大鼠比目鱼肌和趾长伸肌PGC1α/FNDC5/MSTN差异表达但不影响血清Irisin浓度,提示长期抗阻力量训练后机体可能存在复杂的平衡拮抗机制。 Objective To observe the effect of 8-week climbing ladder exercise with load on the ex-pression of proliferator-activated receptor gamma coactivator 1-alpha(PGC1α)/fibronectin type Ⅲ do-main containing 5(FNDC5)/myostatin(MSTN)in soleus and extensor digitorum longus muscle andthe serum concentration of irisin in Sprague-Dawley(SD)rats. Methods Twenty SD rats were random-ly divided into a control(Con,n=10)group and a climbing ladder(Lad,n=10)group. The Lad grouptook an 8-week and once-every-3-day program of resistance exercise using an improved model ofclimbing ladder with load. Then the soleus and extensor digitorum longus muscles were isolated at 48 hafter exercise. The PGC1α/FNDC5/MSTN m RNA and protein expression in the skeleton muscle were de-tected using quantitative PCR and Western blotting respectively. The serum concentration of irisin wasmeasured using enzyme-linked immunosorbent assay. Results Compared with Con group,the expressionof PGC1α in the soleus muscle reduced significantly(P〈0.05),that of FNDC5 reduced but without sig-nificance and that of MSTN increased significantly(P〈0.05)in Lad group. However,the expression ofPGC1α in the extensor digitorum longus muscle increased significantly(P〈0.05),that of FNDC5 in-creased but without significance and that of MSTN decreased significantly(P〈0.05)in Lad group compared with Con group. There were no significant differences in the serum concentration of irisin be-tween the two groups. Conclusion Eight-week climbing ladder exercise with load on rats induces dif-ferential expression of PGC1α/FNDC5/MSTN in soleus and extensor digitorum longus muscle,but doesn't influence the serum concentration of irisin. It is suggested that there may be a complex mechanismof balancing and antagonism in the body after long-term resistance exercises.
