摘要
本文对狗枣猕猴桃叶的多糖进行分离纯化,同时对其抗氧化活性进行研究。先后利用DEAE-Sepharose Fast Flow柱色谱、葡聚糖凝胶Sephadex G-200柱色谱进行分离纯化得到APs-2-1、APs-3-1、APs-3-2三部分均一多糖;利用苯酚-硫酸法测得三部分均一多糖的总糖含量分别为91.26%、90.23%、88.63%,利用硫酸-咔唑法测得糖醛酸的含量分别为16.20%、27.20%、6.90%;此外,还对狗枣猕猴桃叶多糖及所分离的三种均一多糖的总抗氧化能力(FRAP值)、对羟基自由基和DPPH自由基的清除能力进行测定,结果表明所分离的三种多糖及狗枣猕猴桃叶多糖均具有一定的抗氧化能力,但是抗氧化能力均低于Vc。
In this paper,the polysaccharides were separated and purified to obtain three homogeneous polysaccharides. And the total sugar and uronic acid of the three homogeneous polysaccharides were determined. The total sugar was separated using DEAE-Sepharose Fast Flow and Sephadex G-200 column,and to gain three homogeneous polysaccharides,named APs-2-1,APs-3-1,APs-3-2. The total sugar contents of the three homogeneous polysaccharides were 91. 26%,90. 23% and 88. 63% by phenol-sulfuric acid method. The uronic acid content were 16. 20%,27. 20% and 6. 90% by sulfuric-carbazole method. The antioxidant activity of total sugar and the three homogeneous polysaccharides were measured according total antioxidant capacity( FRAP) and the hydroxyl free radical scavenging capacity and DPPH radical scavenging. The experimental results showed that the separation of three homogeneous polysaccharides and total polysaccharide had certain antioxidant capacity,but lower than that of Vc.
