miR-221/222基因家族进化分析及其在猪睾丸组织中的表达变化 被引量:4

摘要 运用生物信息学方法在miRBase中搜索动物miR-221/222基因家族的序列,利用Ensembl数据库信息确定miR-221/222在基因组中的位置,采用MEGA 5.0构建miR-221/222系统进化树;并利用RT-qPCR技术定量检测猪睾丸组织中不同发育时期miR-221/222的表达变化。结果表明:在34种动物中共搜索到59条miR-221/222序列,所有的miR-221/222基因都存在于基因间隔区。进化分析表明,各物种miR-221/222基因家族可能以不同的方式和速度进化而来。RT-qPCR检测结果显示,miR-221/222在猪睾丸组织中不同发育时期表现出相同趋势的表达规律,即出生后其表达水平先升高,生长发育时期降低,随着性成熟又呈现升高趋势。研究结果为深入理解miR-221/222基因家族的作用机制奠定基础,也为后续研究提供参考。 Sequences of miR-221/222 gene family members of different animal species were searched from miRBase database by bioinformatics method.They were located by Ensembl database,and its phylogenetic tree was constructed by MEGA 5.0.In addition,the expression level of miR-221/222 gene was detected in different developmental stages of porcine testicular tissue by real-time fluorescence quantitative PCR.A total of 59 sequences in 34 species were searched and the analysis of their localization showed that they were located on the intergenic region(IGR).Molecular evolution found that miR-221/222 gene families of different species evolved in different ways with different rates.Besides,the similar expression of miR221/222 was detected in different developmental stages of porcine testicular tissue.The level of expression increased after birth,while declined during the growth and development period,and then increased again in sexual maturity period.The results established the base for deeply elucidating the regulatory mechanism of miR-221/222 gene family and could provide information for subsequent experiment research.
