三乙胺对大鼠体质量和肝肾影响的实验研究

Effects of triethylamine on body weight and liver and kidney in rats

摘要 目的探讨三乙胺对大鼠体质量和肝肾的影响。方法用三乙胺对SD雄性大鼠进行亚急性灌胃染毒试验,将清洁级SD雄性大鼠随机分为4组,即对照组和低(25mg/kg)、中(50mg/kg)、高(100mg/kg)剂量3个染毒组,每组8只大鼠。经口灌胃染毒三乙胺1次/d,染毒时间为28d。染毒结束,处死动物,取血清测定天冬氨酸转氨酶、丙氨酸转氨酶、碱性磷酸酶和总蛋白以及肌酐、尿酸和尿素氮。称重肝、肾脏器质量,计算脏器系数,并对肝肾进行病理学检查。结果各剂量组大鼠染毒3周后体质量开始下降,染毒4周后,中、高剂量组血清中天冬氨酸转氨酶、丙氨酸转氨酶和碱性磷酸酶活力升高,高剂量组总蛋白下降,各剂量组肌酐、尿酸和尿素氮升高,与对照组比较差异均有统计学意义(P<0.05);在光学显微镜下观察可见各剂量组实验动物肝细胞出现不同程度的水肿,中、高剂量组可见肝细胞坏死、胞质疏松呈气球样变、胞核萎缩、裂解等现象;各剂量组实验动物肾小球体积增大,中、高剂量组可见肾小管肿胀并有上皮细胞脱落,高剂量组肾脏有明显出血并有炎症细胞浸润。结论经口染毒三乙胺可致雄性大鼠体质量增长受到抑制,并对其肝脏和肾脏产生一定的毒性作用。 Objective To explore the effects of Triethylamine(TEA)by subacute oral administration on body weight and liver and kidney of male rats.Methods Male SD rats were randomly divided into three experimental groups and one control group with 8 rats in each group.The experimental groups were administrated with TEA one time daily for 28 days.At the end of the exposure,animals were sacrificed to measure serum aspartate aminotransferase,alanine aminotransferase,alkaline phosphatase and total protein and creatinine,uric acid and urea nitrogen, weighed liver、kidney organ weight,organ coefficient calculation,pathological examination was performed for liver and kidney.Results The weight of rats in the experimental groups began to decline after the TEA administration for three weeks.The serum aspartate aminotransferase,alanine aminotransferase and alkaline phosphatase increased in medium and high dose groups,total protein decreased in high dose group;the serum creatinine,uric acid and urea nitrogen increased in each dose group,there were significant differences when compared with the control group(P〈0.05).Liver cells were found edema in each dose group,in the medium and the high dose group liver cells showed necrosis,cytoplasm was loose and balloon-like degeneration,the nuclear were pyknosis and karyolysis;glomerular volume increased in each dose group,there was found tubular swelling and epithelial cells detachment in the medium and the high dose groups,and kidneys have significant hemorrhage and inflammatory cellular infiltration in the high dose group.Conclusions The increase inhibition of the body weight and some toxic effects on the liver and kidney were found when the male rats were treated with subacute oral administration of TEA.
