川贝母水提取物对放射性肺炎小鼠模型治疗作用的研究 被引量:10

摘要 目的探讨川贝母水提物对放射性肺炎小鼠的治疗作用。方法 60只C57BL/6小鼠随机分为治疗组、阳性药组、空白组和模型组,治疗组、阳性药组和模型组小鼠每天给予胸部照射(18 Gy),照射后治疗组给予川贝母水提取物(231 mg/kg),阳性药组给予地塞米松(1.5 mg/kg),空白组与模型组则给予等量生理盐水。治疗30 d后处死小鼠,取小鼠肺,部分肺组织制备匀浆,检测其中氧化应激指标(MDA、SOD、GSH-Px),炎性因子(IL-6、TNF-α、IL-12、hs-CRP)含量。其余肺组织行HE及Masson染色,分别观察其肺泡炎发生情况及胶原沉淀情况。结果 HE与Masson染色结果显示,空白组小鼠无肺泡炎及胶原沉积,治疗组小鼠仅出现轻度肺泡炎及胶原沉积,模型组主要为中度及重度肺泡炎及胶原沉积,治疗组小鼠肺泡炎及胶原沉积评分均明显低于模型组。治疗组肺组织I L-6、TNF-α、IL-12、hs-CRP及M DA含量均明显低于模型组,而SOD与GSH-PX明显高于模型组。结论川贝母水提取物治疗放射性肺炎小鼠具有较好的疗效,值得进行深入研究。 Objective To investigate the therapeutic effects oi fritillaria cirrhosa water extraction on the mice with radiation-induced lung injury.Methods Sixty C57BL/6 mice were divided into four groups:treatment group,positive group,control group and model group.The mice in treatment group positive group and model group were exposed to radiation of 18 Gy which was delivered to the thorax.The treatment group and positive group received water extraction oi jritillaria cirrhosa(231 mg/kg) and hexadecadrol(1.5 mg/kg) daily after the radiation,while the control group and model group received solution physiologique.The mice were sacrificed 30 days after radiation and the lungs of mice were fixed and sectioned.The levels of MDA,SOD,GSH-Px,IL-6,TNF-α,IL-12 and hs-CRP in the pulmonary homogenate of mice were detected.The alveolitis and collagen deposition was observed by HE and Masson staining.Results HE and Masson staining showed that the lung was normal in control group and there were mild alveolar inflammatory changes and collagen deposition in treatment group and moderate changes in model group.The scores of HE and Masson in treatment group were lower than those of model group.The levels of MDA,IL-6,TNF-α,IL-12 and hs-CRP in treatment group were lower than those of model group,while the levels of SOD and GSH-PX were higher.Conclusion The water extraction oi fritillaria cirrhosa has good effect on mice with radiation-induced lung injury and worth further research.
