摘要 目的研究脓毒症患者与健康人晚期糖基化终末产物受体(RAGE)多态性分布规律,并探讨与脓毒症的关系。方法选择152例脓毒症患者(脓毒症组)及189例健康体检者(对照组)作为研究对象。应用基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)技术对RAGE基因rs9469089、rs2070600、rs184003、rs2071288多态位点进行检测,并应用酶联免疫吸附法(ELISA)检测两组受试者血浆内皮素1(ET-1)水平,比较两组RAGE基因位点多态性分布规律,探讨其与脓毒症之间的关系。结果脓毒症组患者血浆ET-1水平显著高于对照组,差异有统计学意义(P<0.05)。脓毒症组与对照组RAGE rs9469089、rs2070600、rs184003、rs2071288多态性位点的等位基因及基因型分布频率差异均无统计学意义(P>0.05)。结论脓毒症患者血浆ET-1表达水平显著升高,所检测的RAGE基因多态性与脓毒症的易感性无相关。 Objective To study the polymorphism distribution of the receptor for advanced glycation end products(RAGE)in patients with sepsis and in healthy people,and to explore the possible relationship between the polymorphism of the receptor and the sepsis. Methods A total of 152 cases of sepsis(sepsis group)and 189 cases of healthy subjects(control group)were selected in this study.The RAGE gene polymorphisms of loci rs9469089,rs2070600,rs184003 and rs2071288were detected by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF-MS).The levels of plasma endothelin-1(ET-1)in the two groups were measured by ELISA.The distribution of RAGE gene polymorphism was compared between the two groups and the relationship between the RAGE gene polymorphism and sepsis was analyzed. Results Plasma ET-1levels in sepsis group were significantly higher than those in control group,and there was statistical difference(P〈0.05).The allele frequency and genotype frequency of RAGE rs9469089,rs2070600,rs184003,and rs2071288 polymorphism were not significantly different between the sepsis group and the control group,and there was no statistical difference(P〉0.05).Conclusion Serum ET-1levels are higher in patients with sepsis.RAGE gene polymorphism may not be associated with the susceptibility to sepsis.
