腺苷A1受体信号通路在七氟醚后处理减轻大鼠局灶性脑缺血再灌注损伤中的作用 被引量:1

Role of Adenosine A1 Receptor Signal Pathway in Alleviating Focal Cerebral Ischemia Reperfusion Injury after Sevoflurane Treatment in Rats

摘要 目的 评价腺苷A1受体信号通路在七氟醚后处理减轻大鼠局灶性脑缺血再灌注损伤中的作用.方法 健康雄性成年SD大鼠85只,体重250-280 g,随机分为5组(每组17只):假手术组(Sham组)、缺血再灌注组(I/R组)、七氟醚后处理组(Se组)、七氟醚后处理+腺苷A1受体拮抗剂1,3-二丙基-8-环戊基腺嘌呤(DPCPX)组(Se+D组)和拮抗剂DPCPX组(D组).采用大脑中动脉栓塞(MCAO)法制备局灶性脑缺血模型.再灌注即刻Se组和Se+D组吸入2.4%的七氟醚60 min,I/R组和D组吸入纯氧.脑缺血前30 min时Se+D组和D组腹腔注射DPCPX 1 mg/kg.于再灌注6 h时行神经功能评分,再灌注72 h时取脑组织,2,3,5-氯化三苯基四氮唑(2,3,5-triphenyltetrazolium chloride,TTC)染色测定脑梗死体积百分比.于再灌注24 h时取脑组织,观察脑组织的病理学结构,酶联免疫吸附法(ElISA)测定TNF-α,IL-1β的浓度.结果 与Sham组比较,I/R组,Se组,Se+D组,D组神经功能评分升高,脑梗死体积百分比增加,缺血区脑组织TNF-α,IL-1β的含量明显增加(P〈0.05);与I/R组,Se+D组,D组比较,Se组神经功能评分降低,脑梗死体积百分比增加,缺血区脑组织TNF-α,IL-1β的含量明显降低(P〈0.05).结论 腺苷A1受体信号通路的激活,可能参与了七氟醚后处理减轻大鼠局灶性脑缺血再灌注损伤的过程. Objective To investigate the role of adenosine A1 receptor signal pathway in alleviating focal cere- bral ischemia reperfusion injury after sevoflurane treatment in rats. Methods 85 adult male SD rats weighing 250 280 g were randomly divided into five groups with 17 rats in each group : sham operation group ( group sham), focal cerebral I/R group( group I/R) ,sevoilurane treatment group( group Se) ,sevoflurane treatment + adenosine A1 recep- tor inhibitor 8 - Cyclopentyl - 1,3 - dipropylxanthine(DPCPX) group( group S + D) and DPCPX group( group D). Fo- cal cerebral I/R model was established by middle cerebral artery occlusion(MCAO). Rats in group Se and group Se + D inhaled 2.4% sevoflurane for 60 min, and rats in group I/R and group D inhaled pure oxygen. DPCPX 1 mg/kg was injected intraperitoneally 30 min before ischemia. Neurological deficit scores (NDSs) were assessed 6 h of reperfusion. Rat brain tissue was removed 72 h after reperfusion. Percentage of cerebral infarction volume was measured by 2,3,5 - triphenyltetrazolium chloride. Rat brain tissue was removed 24 h after reperfusion. Pathology structure of brain tis- sue was observed and concentrations of TNF - ct and IL - 113 were detected by enzyme linked immunosorbent assay (ELISA). Results NDSs, percentage of cerebral infarction volume as well as concentrations of TNF - a and IL - 113 were significantly higher in the other 4 groups than those in group sham(P 〈0.05). NDSs were lower,percentage of cerebral infarction volume was higher and concentrations of TNF - o~ and IL - 113 were lower in group Se than those in group I/R, group S + D and group D ( P 〈 0.05 ). Conclusion Adenosine A1 receptor signal pathway may participate in alleviating focal cerebral ischemia reperfusion injury after sevoflurane treatment in rats.
